白内障晶状体上皮细胞中miR-126和VEGF的表达关系及意义

摘　　要：目的探讨白内障晶状体上皮细胞中miR-126和VEGF的表达关系及意义。方法选择白内障患者198例为病例组,正常晶状体前囊膜标本49例为对照组。测定2组白内障晶状体上皮细胞中miR-126、VEGF水平;同时设SRA01/04细胞组、MIR-126 inhibitor、MIR-126 mimics组,测定3组SRA01/04细胞OD值、存活率、凋亡率、G1期、MIR-126、VEGF mRNA、Caspase-3、Caspase-6、Caspase-9蛋白表达水平。结果病例组MIR-126、VEGF mRNA水平高于对照组(P<0.01);MIR-126 mimics组OD值、存活率水平低于SRA01/04细胞组(P<0.05),MIR-126 inhibitor组OD值、存活率水平高于SRA01/04细胞组、MIR-126 mimics组(P<0.05);MIR-126 mimics组凋亡率水平高于SRA01/04细胞组(P<0.05),MIR-126 inhibitor组凋亡率水平低于SRA01/04细胞组、MIR-126 mimics组(P<0.05);MIR-126 mimics组G1期水平低于SRA01/04细胞组(P<0.05),MIR-126 inhibitor组G1期水平高于SRA01/04细胞组、MIR-126mimics组(P<0.05);MIR-126 mimics组MIR-126、VEGF mRNA水平高于SRA01/04细胞组(P<0.05),MIR-126 inhibitor组MIR-126 mRNA、VEGF mRNA水平低于SRA01/04细胞组、MIR-126 mimics组(P<0.05);MIR-126 mimics组Caspase-3、Caspase-6、Caspase-9蛋白表达水平高于SRA01/04细胞组(P<0.05),MIR-126 inhibitor组Caspase-3、Caspase-6、Caspase-9蛋白表达水平低于SRA01/04细胞组、MIR-126 mimics组(P<0.05);MIR-126 mRNA与VEGF、Caspase-3、Caspase-6、Caspase-9正相关关系明显(P<0.05)。结论miR-126、VEGF在白内障晶状体上皮细胞中高表达;MIR-126能诱导白内障晶状体上皮细胞高表达VEGF、Caspase-3、Caspase-6、Caspase-9,进而促进其凋亡。Objective To investigate the expression and significance of microRNA-126(miR-126)and vascular endothelial growth factor(VEGF)in lens epithelial cells of patients with cataract.Methods A total of 198 patients with cataract were enrolled as case group,and 49 cases of normal anterior capsule specimens were served as control group.The expression levels of miR-126 and VEGF in cataract lens epithelial cells were detected in both groups.Meanwhile,SRA01/04 cell group,MIR-126 inhibitor group,MIR-126 mimics group were set up,and the OD value of SRA01/04 cells,survival rate,apoptosis rate,cells numbers at G1 phase,mRNA expression levels of MIR-126 and VEGF,protein expression levels of Caspase-3,Caspase-6 and Caspase-9 were detected.Results The expression levels of MIR-126,VEGF mRNA in case group were significantly higher than those in control group(P<0.01).The OD value and survival rate in MIR-126 mimics group were significantly lower than those in SRA01/04 cell group(P<0.05),however,which in MIR-126 inhibitor group were significantly higher than those in SRA01/04 cell group(P<0.05).The cell apoptosis rates in MIR-126 inhibitor group were significantly lower than those in SRA01/04 cell group and those in MIR-126 mimics group(P<0.05).Moreover the cell levels at Gi stage in MIR-126 mimics group were significantly lower than those in SRA01/04 cell group(P<0.05),which in MIR-126 inhibitor group were significantly higher than those in SRA01/04 cell group and those in MIR-126 mimics group(P<0.05).The expression levels of MIR-126,VEGF mRNA in MIR-126 mimics group were significantly higher than those in SRA01/04 cell group(P<0.05),however,which in MIR-126 inhibitor group were significantly lower than those in SRA01/04 cell group and those in MIR-126 mimics group(P<0.05).In addition the expression levels of Caspase-3,Caspase-6,Caspase-9 were significantly higher than those in SRA01/04 cell group,which in MIR-126 inhibitor group were significantly lower than those in SRA01/04 cell group and those in MIR-126 mimics group(P<0.05).Besid
