摘 要:研究影响白及多糖提取的因素,优化其提取工艺及探究其抑菌活性。通过单因素和响应曲面法,选择提取次数、水提时间、水提温度、液料比4个因素筛选白及多糖的最佳水提工艺,蒽酮-硫酸法测定白及多糖的含量,滤纸片-抑菌圈法测定白及多糖对大肠杆菌、金黄色葡萄球菌、枯草芽孢杆菌及黑曲霉菌的抑菌作用。经由以上实验设计可得白及多糖最佳提取工艺为:提取次数1次、水提时间2.2 h、水提温度77℃、液料比39.27 mL/g,白及多糖得率31.09%;白及多糖对大肠杆菌、金黄色葡萄球菌、枯草芽孢杆菌及黑曲霉菌均有抑制作用。To study the factors affecting the extraction of Bletilla Striata polysaccharide,optimize the extraction process and explore its antibacterial activity.By single factor and response surface method,four factors including extraction times,water extraction time,water extraction temperature,liquid-material ratio were selected the best water extraction process for Bletilla Striata polysaccharide,anthrone-sulfuricacid method to determine the content ofBletilla Striata polysaccharide,bacteriostatic circle a method to determine Bletilla Striata polysaccharide in e.coli,staphylococcus aureus,bacillus subtilis and aspergillus bacteriostatic action.According to the above experimental design,the optimal extraction process of Bletilla Striata polysaccharide was as follows:extraction times 1,waterextraction temperature 77℃,water extraction time 2.2h,liquid-solid ratio 39.27mL/g,and yield of Bletilla Striata polysaccharide 31.09%.Bletilla Striata polysaccharide can inhibit Escherichia coli,Staphylococcus aureus,Bacillus subtilis andAspergillus Niger.
