摘 要:目的探讨肿瘤抑制因子B细胞易位基因1(BTG1)与编码转录因子蛋白的基因1(KZF1)在小鼠白血病发生过程中的相互作用。方法将24只SPF级BALB/c裸鼠分为对照组和模型组,每组12只。模型组采用尾静脉注射白血病细胞系K562建立白血病模型,对照组注射等剂量生理盐水。试验后1周、3周与6周采用MTT法检测细胞生存率,酶联免疫法检测肿瘤坏死因子α(TNF-α)与白细胞介素6(IL-6)含量;试验后6周流式细胞仪法检测细胞凋亡,Western blot法检测BTG1、IKZF1蛋白表达水平;定量分析小鼠肝和肺中白细胞浸入情况。结果试验后1周、3周与6周,模型组的造血细胞生存率、血清TNF-α与IL-6含量显著高于对照组(P<0.05),模型组在组内对比差异也有统计学意义(P<0.05)。试验后6周,模型组的骨髓细胞凋亡指数显著高于对照组(P<0.05),骨髓BTG1、IKZF1蛋白相对表达水平显著低于对照组(P<0.05)。与对照组相比,模型组渗入小鼠肝和肺的白细胞百分比显著上升(P<0.05)。结论小鼠白血病发生过程伴随有BTG1和IKZF1的低表达,可促进炎症因子的释放,导致造血细胞增殖旺盛与骨髓细胞凋亡并使得小鼠肝和肺组织的白细胞百分比上升。Objective To investigate the interaction between BTG1 and IKZF1 in mouse leukemia.Methods 24 cases of SPF-level BALB/c nude mice were divided into control group and model group,with 12 mice in each group.The model group were performed the tail vein injection with leukemia cell line K562 to establish leukemia model,the control group were injected with equal dose of normal saline.Cell viability were detected by MTT assay,TNF-αand IL-6 levels were detected by enzyme-linked immunosorbent assay,apoptosis were detected by flow cytometry,and BTG1 and IKZF1 protein expression levels were detected by Western blot.Leukocyte infiltration in liver and lung of mice was quantitatively analyzed.Results After 1,3 and 6 weeks after the treatment,the hematopoietic cell survival rates,serum TNF-αand IL-6 levels in the model group were significantly higher than those in the control group(P<0.05),and theere were also statistically significant difference in the model group compared in the different times(P<0.05).At 6 weeks after the experiment,the apoptotic index of bone marrow cells in the model group were significantly higher than that in the control group(P<0.05),and the relative expression levels of BTG1 and IKZF1 proteins in the bone marrow were significantly lower than those in the control group(P<0.05).Compared with the control group,the percentage of leukocytes that infiltrated into the liver and lung of mice increased significantly in the model group(P<0.05).Conclusion The development of mouse leukemia are accompanied by the low expression of BTG1 and IKZF1,which can promote the release of inflammatory factors,leading to the proliferation of hematopoietic cells and apoptosis of bone marrow cells and increases the percentage of white blood cells in mouse liver and lung tissue.
