摘 要:目的:观察黄芪甲苷对人内皮祖细胞(endothelial progenitor cells,EPCs)分泌血管生长因子的影响,分析黄芪甲苷调节EPCs介导的血管新生的作用机制。方法:取足月健康新生儿脐带血,用密度梯度离心法获取单个核细胞,进行传代培养,当细胞呈现梭形时对细胞进行CD31抗体和DAPI核染鉴定,获得EPCs。将获得的EPCs随机分为实验组和对照组,实验组用质量浓度为100 mg L-1的黄芪甲苷干预,对照组用等量的PBS液处理,培养24 h后,通过酶联免疫吸附实验检测血管内皮生长因子(vascular endothelial growth factor,VEGF)a、VEGFb、VEGFc、成纤维细胞生长因子(fibroblast growth factors,FGF)、血管生成素-1(angiogenin-1,Ang-1)的吸光度。结果:经CD31抗体和DAPI核染检测证实成功获取EPCs。实验组人EPCs分泌的血管生长因子VEGFa、VEGFb、VEGFc、FGF、Ang-1水平均高于对照组(分别t=9.84、45.24、14.51、6.27、6.57),差异均有统计学意义(P<0.01)。结论:黄芪甲苷可促进人EPCs分泌大量VEGF,具有调节EPCs介导血管新生的巨大潜能。Objective:To observe the effect of astragaloside on the secretion of vascular growth factors by human endothelial progenitor cells(EPCs),and to study the mechanism of astragaloside in regulating EPCs-mediated angiogenesis.Methods:Umbilical cord blood of full-term healthy neonates was collected,and mononuclear cells were obtained by density gradient centrifugation and subcultured.When the cells showed a spindle shape,the cells were identified by CD31 antibody and DAPI nuclear staining to obtain EPCs.The obtained EPCs were randomly divided into an experimental group and a control group.The experimental group was intervened with astragaloside at a concentration of 100 mg L-1,and the control group was treated with an equal amount of PBS solution.After 24 hours of incubation,vascular endothelial growth factor(VEGF)a,VEGFb,VEGFc,fibroblast growth factors(FGF),and angiogenin-1(Ang-1)were detected by enzyme-linked immunosorbent assay angiogenin-1(Ang-1)absorbance.Results:The EPCs were successfully obtained by CD31 antibody and DAPI nuclear staining test.The vascular growth factors VEGFa,VEGFb,VEGFc,FGF and Ang-1 secreted by human EPCs in the experimental group were higher than those in the control group(t=9.84,t=45.24,t=14.51,t=6.27,t=6.57,respectively).The differences are statistically significant(P<0.01).Conclusion:Astragaloside can promote the secretion of large amounts of VEGF by human EPCs,and has great potential to regulate EPCsmediated angiogenesis.
