摘 要:目的探讨宫颈癌细胞株中膜联蛋白A7(Annexin A7)表达抑制后对Ki67和P16的影响。方法细胞培养后分成siRNA干扰组、阴性对照组和空白对照组进行小干扰RNA(siRNA)干扰,抑制宫颈癌细胞系Hela细胞Annexin A7的表达,蛋白印迹法鉴定其干扰效率后,应用激光共聚焦免疫荧光法检测Annexin A7表达抑制后对Hela细胞Ki67和P16表达的影响。结果干扰组Annexin A7的表达量与阴性对照组和空白对照组比较明显降低,差异有统计学意义(P<0.05);与阴性对照和空白对照组比较,Ki67阳性产物在siRNA干扰组明显减少,差异有统计学意义(P<0.05);siRNA干扰组、阴性对照组和空白对照组P16阳性产物比较差异无统计学意义(P>0.05)。结论Annexin A7可能参与了宫颈癌细胞增殖功能的上调作用。Objective To study the effect of Annexin A7 expression inhibition on Ki67 and P16 of Hela cells.Methods After cell culture,the cells were divided into the siRNA interference group,the negative control group and the blank control group for small interfering RNA(siRNA)interference to inhibit the expression of Annexin A7 in Hela cells of cervical cancer cell line.After the interference efficiency was identified by Western blot,the effect of the inhibition of Annexin A7 on the expression of Ki67 and P16 in Hela cells was detected by laser confocal immunofluorescence assay.Results Annexin A7 expression in the siRNA intervention group was significantly lower than that in the negative control group and the blank control group,with statistically significant difference(P<0.05).Compared with the negative control group and the blank control group,Ki67 positive products were significantly reduced in the siRNA interference group,and the difference was statistically significant(P<0.05).There was no statistically significant difference in P16 positive products among the siRNA interference group,the negative control group and the blank control group(P>0.05).Conclusion Annexin A7 may get involved in upregulated cell proliferation of squamous cell carcinoma of the cervix.
