摘  要:目的:探讨二甲双胍和阿霉素联合用药对弥漫大B细胞淋巴瘤的影响,及其作用机制。方法:MTS检测细胞的增殖能力;流式细胞术分析药物干预DLBCL细胞凋亡及周期变化;蛋白免疫印迹法(Western blot)检测PI3K、AKT、Caspase3、Cytochrome=C蛋白表达。结果:MTS结果显示两个药物联合处理对细胞的抑制率显著高于各药物单独处理组。相对control组,各药物处理组的细胞凋亡显著增加(P<0.05),并且各药物单独处理组细胞的凋亡比率显著低于联合处理组(P<0.05)。相比control组,各药物处理组PI3K和AKT蛋白表达显著降低(P<0.05),凋亡相关蛋白Caspase3和Cytochrome C表达显著升高(P<0.05),并且联合处理组Caspase3、Cytochrome C表达显著高于各药物单独处理组(P<0.05)结论:两个药物联合使用通过PI3K/AKT信号通路影响细胞增殖与凋亡,从而增加了阿霉素对细胞的毒性作用。Objective:To investigate the effect of metformin and doxorubicin on diffuse large B-cell lymphoma and its mechanism of action.Methods:MTS method was used to detect the cell proliferation ability;flow cytometry method was used to analyze the drug intervention for DLBCL cell apoptosis and cycle changes;Western blot method was used to detect PI3K,AKT,Caspase3,Cytochrome C protein expression.Results:MTS detection results showed that the inhibition rate of two drugs combined treatment on cells was significantly higher than that of each drug treatment group alone.Compared with the control group,the apoptosis of each drug treatment group increased significantly(P<0.05),and the apoptosis rate of the cells treated with each drug alone was significantly lower than that of the combined treatment group(P<0.05).Compared with the control group,the expressions of PI3K and AKT proteins in each drug treatment group were significantly reduced(P<0.05),the expressions of apoptosis-related proteins Caspase3 and Cytochrome C were significantly increased(P<0.05),and the expressions of Caspase3 and Cytochrome C were significantly higher in the combined treatment group.Higher than each drug treatment group(P<0.05).Conclusion:The combined use of the two drugs affects cell proliferation and apoptosis through the PI3K/AKT signaling pathway,thereby increasing the toxic effect of adriamycin on cells.
