摘  要:目的:检测Tim-3-HMGB1在HBV相关肝癌患者中的表达水平.方法:收集35例肝癌患者及40例健康对照外周血,采用Ficoll-Hypaque梯度离心法分离外周血单个核细胞(PBMC),分别提取血清病毒DNA及总RNA,普通聚合酶链反应(PCR)检测HBV-DNA、Tim3表达水平;实时荧光定量PCR检测HMGB1表达水平.结果:HBV相关肝癌患者外周血单个核细胞Tim-3(P<0.01)-HMGB1(P=0.019)表达水平均增高.结论:HBV相关肝癌患者外周血单个核细胞Tim-3-HMGB1表达水平增高,可作为辅助诊断指标应用.Objective:To detect the expression of tim-3-HMGB1 in patients with HBV related hepatocellular carcinoma(HCC).Methods:The peripheral blood of 35 patients with liver cancer and 40 healthy controls were collected.Peripheral blood mononuclear cells(PBMC)were isolated by Ficoll Hypaque gradient centrifugation,and virus DNA and total RNA were extracted respectively.HBV-DNA and tim3 expression levels were detected by polymerase chain reaction(PCR).HMGB1 expression was detected by real-time fluorescence quantitative PCR.Results:the mRNA expression level of Tim-3(P<0.01)-HMGB1(P=0.019)in PBMCs of patients with HBV related liver cancer were increased.Conclusion:The expression of tim-3-HMGB1 in PBMCs of patients with HBV related liver cancer could be used as an auxiliary diagnostic index.
