摘 要:目的观察加减葛花解酲汤预处理对急性酒精中毒小鼠的肝脏保护作用,并探讨其作用机制。方法将40只昆明雄性小鼠随机分为对照组、模型组及加减葛花解酲汤低、中、高剂量组各8只。模型组给予16 mL/kg白酒灌胃,建立急性酒精中毒模型;低、中、高剂量组先给予0.5、1、2 g/mL加减葛花解酲汤20 mL/kg灌胃,30 min后再给予16 mL/kg白酒灌胃;对照组给予等量蒸馏水灌胃。灌胃结束后,检测血液中的乙醇含量;取肝脏组织,采用ELISA法检测乙醇代谢相关酶乙醇脱氢酶(ADH)、乙醛脱氢酶(ALDH)及肝功能指标ALT、AST活性。结果加减葛花解酲汤低、中、高剂量组血液中的乙醇含量均低于模型组(P均<0.05)。与对照组比较,模型组ADH、ALDH、ALT、AST活性均降低(P均<0.05)。与模型组比较,低、中剂量组ADH活性升高(P均<0.05),高剂量组无明显变化(P>0.05);低、中、高剂量组ALDH、ALT、AST活性均高于模型组(P<0.05或<0.01)。结论加减葛花解酲汤预处理可降低急性酒精中毒小鼠血液中的乙醇含量,提高肝组织中乙醇代谢酶的活性,促进乙醇代谢,从而发挥肝脏保护作用。Objective To observe the protective effect of modified Gehua Jiecheng decoction on the liver of acute alcoholism mice and to explore its mechanism.Methods Forty male Kunming mice were randomly divided into the control group,model group,and low-dose,medium-dose and high-dose modified Gehua Jiecheng decoction groups with 8 in each.The mice in the model group were given 16 mL/kg liquor to establish the acute alcoholism models,the low-dose,medium-dose and high-dose modified Gehua Jiecheng decoction groups were first given 0.5,1,2 g/mL modified Gehua Jiecheng decoction of 20 mL/kg,and 30 min later,they were given 16 mL/kg liquor,and the control group were administered equal amounts of distilled water to the stomach.The concentration of ethanol in blood was measured after the gastric perfusion.The activities of ethanol metabolic-related enzymes,ethanol dehydrogenase(ADH),acetaldehy dehydrogenase(ALDH),alanine transaminase(ALT),and aspartate transaminase(AST)were detected by ELISA.Results The concentration of alcohol in the blood of the low-dose,medium-dose,and high-dose groups of modified Gehua Jiecheng decoction was lower than that of the model group(all P<0.05).Compared with the control group,the activities of ADH,ALDH,ALT and AST in the model group decreased(all P<0.05).Compared with the model group,the ADH activity in low-dose and medium-dose groups increased(both P<0.05),and there was no significant change in the high-dose modified Gehua Jiecheng decoction group(P>0.05).The activities of ALDH,ALT and AST in the low-dose,medium-dose and high-dose modified Gehua Jiecheng decoction groups were higher than that in the model group(P<0.05 or P<0.01).Conclusion The pretreatment of modified Gehua Jiecheng decoction can reduce the concentration of ethanol in the blood of mice with acute alcoholism,improve the activity of ethanol metabolic enzymes in the liver tissues,promote ethanol metabolism,and thus protect the liver.
