摘  要:目的制备活性多肽奶基质脂质体,对其物理化学性质进行表征并添加于凝胶、溶液和乳液中,制备脂质体制剂。方法以牛奶、豆奶等奶基质采用薄膜分散法制备脂质体;采用激光粒度仪考察脂质体粒径与电位的分布情况;设计不同的防腐配比筛选脂质体凝胶制剂最优防腐方案;于4℃和室温条件下考察制剂稳定性。结果2种奶基质脂质体粒径约150 nm,多分散系数均<0.2,表明分散度良好;脂质体电位约-20 mV,有利于分散稳定。防腐剂α-红没药醇和ε-聚赖氨酸的含量分别为0.5%和0.3%时脂质体凝胶制剂具有优异的防腐性能。将脂质体添加于凝胶、溶液和乳液中,所得脂质体制剂外观均匀分散,稳定性良好,其中所制得的活性多肽奶基质脂质体胶体制剂具有胶体系统所特有的蓝色乳光和丁达尔效应。结论以牛奶、豆奶等奶基质,采用薄膜分散法制备的脂质体,稳定性好,方法简便易行,添加于各种皮肤外用制剂中,用于递送活性物质,改善皮肤状态。Objective To prepare the active polypeptide milk-matrix liposomes,characterize their physicochemical proper ties,and add the liposomes into gels,solutions and emulsions to prepare liposomal preparation.Methods Liposomes were prepared using milk,soymilk and other milk as matrixes by film dispersion method.The distribution of particle size and potential of liposomes were investigated by laser particle size analyzer.Different preservatives and their ratios were tested to obtain optimal anti-corrosion measures for liposomal gel preparation.The stability of preparations was investigated at 4℃ and room temperature.Results The parti cle size of the two milk-matrix liposomes was around 150 nm,and the polydispersity index was less than 0.2,which indicated that the dispersity of the liposomes was good.The potential of the liposomes was around-20 mV,which was conducive to the dispersion and stability.The liposomal gel preparation showed excellent antiseptic properties when the 0.5%and 0.3% α-bisabolol andε-polylysine were used as the preservatives,respectively.The liposomal preparations,prepared by adding the liposome to the gels,solutions and emulsions,appeared to be uniformly dispersed and showed a good stability.The prepared active polypeptide milk-matrix liposome col loid preparations showed the colloidal system-characteristic blue opalescence and Tindal effect.Conclusion The aimed liposomes could be simply prepared by the film dispersion method using milk and soymilk etc as matrixes and showed a good stability,which might be added to various skin external preparations to deliver active substances to improve skin conditions.
