摘  要:目的通过在颈背部皮下注射鱼藤酮试剂进行造模,建立帕金森病模型。方法将45只SPF级SD雄性大鼠随机分为正常组、假手术组、模型组,模型组给予颈背部皮下注射鱼藤酮试剂进行造模,假手术组仅注射等量葵花油乳化液(不含鱼藤酮),正常组不处理。观察其外观形态学改变,进行行为学测试、旷场实验、悬挂实验、滚轴实验并进行评分。免疫组织化学法检测大鼠黑质内α-突触核蛋白(α-synuclein,α-syn)与酪氨酸羟化酶(Tyrosine hydroxylase,TH)阳性表达。结果(1)模型组大鼠出现弓背姿势,毛色变黄,背毛变硬呈扇形等改变。模型组大鼠行为学测试评分明显提高(P<0.01);旷场实验、悬挂实验、滚轴实验结果均显示模型组大鼠与正常组和假手术组相比行为异常(P<0.01)。(2)免疫组化检测结果显示模型组大鼠黑质内α-syn平均光密度值显著增高(P<0.01),TH平均光密度值显著降低(P<0.01)。结论用颈背部皮下注射鱼藤酮试剂的方法可建立与人类帕金森病行为和病理高度相似的大鼠模型,为PD实验的模型建立提供理论基础。Objective To establish a model of Parkinson’s disease by subcutaneous injection of rotenone in the back of the neck. Methods Forty-five SPF SD male rats were randomly divided into normal group, sham operation group and model group. The model group was given subcutaneous injection of rotenone reagent for modeling. The sham operation group only injected the same amount of sunflower oil emulsion(without rotenone), no treatment in the normal group. Morphological changes in appearance were observed, and behavioral tests, open field experiments, suspension experiments, roller experiments were performed and scored. The expressions of α-synuclein(α-syn) and Tyrosine hydroxylase(TH) in the substantia nigra of rats were detected by immunohistochemistry.Results(1)The model group showed a bow-back posture, the hair color turned yellow, and the back hair became hard and fan-shaped. The behavioral test scores of the model group were significantly improved(P<0.01). The results of open field experiment, suspension experiment and roller test showed that the rats in the model group had abnormal behavior compared with the normal group and the sham operation group(P<0.01).(2)Immunohistochemical results showed that the mean optical density of α-syn in the substantia nigra of the model group was significantly increased(P<0.01), and the average optical density of TH was significantly decreased(P<0.01). Conclusion The method of injecting rotenonereagent subcutaneously under the neckcan establish a rat model that is highly similar to human Parkinson’s disease behavior and pathology, and provide a theoretical basis for model establishment of PD experiments.
