摘  要:目的探讨不同介入时间高压氧(HBO)治疗对脑出血大鼠出血灶周围GAP-43表达的影响。方法应用胶原酶诱导法建立大鼠脑出血模型,将185只雄性Wistar大鼠随机分为正常组、假手术组、脑出血对照组和HBO治疗组,其中,正常组包含5只大鼠,其余三组各有60只大鼠。HBO治疗组按HBO介入时间不同分为四个亚组:6h介入组(15只),1d介入组(15只),2d入组(15只)和3d介入组(15只),再按不同治疗次数分为三个亚组,分别为:HBO后1d组(5只),3d组(5只)和5d组(5只),治疗方案为:HBO治疗压力2.0ATA,稳压吸氧60min,1次/d。所有实验大鼠饲养在相同环境中,正常组饲养3d后处死,脑出血对照组与假手术组大鼠不进行任何治疗,参照HBO组进行分组,并于相应时间点断头取脑,采用免疫组化法测定GAP-43的表达。结果①HBO治疗组及脑出血对照组大鼠出血灶周围GAP-43的表达均高于正常组及假手术组(P<0.05);②HBO治疗组较相应对照组大鼠出血灶周围GAP-43表达增加(P<0.05);③HBO治疗组中介入时间为6h的大鼠出血灶周围GAP-43表达较其他对应治疗组增加(P<0.05)。结论HBO治疗促进实验性脑出血大鼠出血灶周围GAP-43表达水平增加,且早期介入HBO治疗效果较好。Objective To observe the effect of hyperbaric oxygen(HBO)treatment with different intervention time on the expression of growth-related protein-43 around the hemorrhagic brain tissue in experimental cerebral hemorrhage rats.Methods The rat model of cerebral hemorrhage was established by collagenase induction.185 male Wistar rats were randomly divided into normal group(5 rats),sham-operated group,cerebral hemorrhage control group and HBO therapy group.The last three groups have 60 rats separately.The HBO therapy group was divided into four subgroups:6 h intervention group,1 d intervention group,2 d group and 3 d intervention group,on average 15 rats in each group.The number of times was divided into 1 d,3 d,5 d group of HBO treatment,5 rats at each time point.The HBO treatment pressure is 2.0 ATA,with the duration of 60 minutes steady oxygen inhalation every day.The rats were all killed after 24 hours at the last time of HBO treatment.All the rats were adopted in the same environment,and the normal group was sacrificed after 3 days.The cerebral hemorrhage control group and the sham-operated group did not receive any treatment.The rats were grouped like the HBO treatment group.The brains were decapitated at the corresponding time points.We detected the expression of GAP-43 using the way of immunohistochemistry.Results The expression of GAP-43 around the hemorrhage in the HBO therapy group and the cerebral hemorrhage control group was higher than that in the normal group and the sham operation group(P<0.05);The expression of GAP-43 was increased compared with the control group of cerebral hemorrhage(P<0.05);The expression of Gap-43 around the hemorrhage of the rats in 6 h group was higher than that of the other three groups in the HBO treatment group(P<0.05).Conclusion HBO treatment can increase the expression of GAP-43 around the hemorrhage brain tissue in rats with experimental cerebral hemorrhage and the optimal intervention time is 6 h.
