摘　　要：目的探讨结直肠癌中E2F-1的表达状态和对肿瘤预后的可能影响。方法收集结直肠癌组织与癌旁正常组织,使用Western blot技术检测两组间E2F-1表达差异。构建E2F-1真核质粒,转染结肠癌LoVo细胞株,通过克隆形成实验和绘制细胞生长曲线,分析E2F-1对结肠癌细胞的影响。结果肿瘤组E2F-1的蛋白表达量高于癌旁对照组,差异有统计学意义(P<0.05)。E2F-1过表达能抑制结肠癌LoVo细胞株的增殖。结论E2F-1在结直肠肿瘤细胞中过度表达,并在体外实验中抑制结肠癌细胞株的增殖。Objective To investigate the expression of E2F-1 in colorectal cancer and its possible influence on tumor prognosis.Methods Collecting colorectal cancer tissue and adjacent normal tissue,and western blot was used to detect the difference of E2F-1 expression between the two groups.The eukaryon of E2F-1 was constructed and transfected into LoVo cell line of colon cancer.The effect of E2F-1 on colon cancer cells was analyzed by clone formation experiment and cell growth curve drawing.Results the protein expression of E2F-1 in tumor group was significantly higher than that in control group(P<0.05).Overexpression of E2F-1 can inhibit the proliferation of LoVo cell line.Conclusion E2F-1 is overexpressed in colorectal cancer cells and inhibits the proliferation of colon cancer cells in vitro.
