摘  要:目的研究人参皂苷Rg3联合沉默MMP-9基因对乳腺癌肿瘤生长和转移的作用,为探讨新的乳腺癌治疗方式和潜在靶点提供理论基础。方法培养高转移性乳腺癌细胞系MDA-MB-231,使用Rg3处理细胞,并且转染MMP-9过表达载体或MMP-9 siRNA沉默载体。细胞划痕实验检测细胞迁移能力变化、采用Transwell小室使用检测细胞侵袭能力、CCK-8检测细胞增值活性、DNA变化检测细胞周期。RT-qPCR检测已处理并转染的MDA-MB-231中MMP-9表达变化,Western blot检测细胞中Ki-67、CDK1、VEGF、Twist1、MMP-9蛋白的表达变化。结果Rg3抑制MDA-MB-231细胞的侵袭和迁移能力,且下调MMP-9的mRNA及蛋白表达(P<0.05),且MMP-9过表达显著逆转Rg3对细胞的侵袭和迁移能力的抑制作用(P<0.05)。Rg3联合沉默MMP-9处理细胞显著抑制高转移性乳腺癌细胞的增殖活性和G1/S周期进展以及Ki-67和CDK1蛋白的表达水平(P<0.05)。Rg3联合沉默MMP-9处理细胞抑制该高转移性乳腺癌细胞的迁移和侵袭能力以及VEGF和Twist1蛋白的表达(P<0.05)。结论人参皂苷Rg3通过抑制MMP-9抑制高转移性乳腺癌细胞的侵袭和迁移,人参皂苷Rg3联合沉默MMP-9抑制乳腺癌肿瘤细胞的生长,迁移及侵袭。Objective To investigate the effects of ginsenoside Rg3 and MMP-9 combined with the silencing of MMP-9 on the growth and metastasis of breast cancer in vitro,so as to provide theoretical basis for exploring new treatment methods and potential targets for breast cancer.Methods The high metastatic breast cancer cell line MDA-MB-231 was cultured in vitro,and the cells were treated with Rg3 and transfected with MMP-9 over-expression vectors or MMP-9 siRNA silencing vectors.The changes of cell migration ability were detected by cell wound healing test,and cell invasion ability was detected by Transwell chamber,cell proliferation activity was detected by CCK-8,and cell cycle was analyzed according to DNA changes.The expression levels of MMP-9 mRNA in the cells were detected by RT-qPCR.Moreover the expression levels of Ki-67,CDK1,VEGF,Twist1,and MMP-9 were detected by Western Blot.Results The Rg3 inhibited the invasion and migration of MDA-MB-231 cells and down-regulated the expression levels of MMP-9 mRNA and protein significantly(P<0.05).In addition MMP-9 over-expression significantly reversed the inhibition of Rg3 on cell invasion and migration(P<0.05).The Rg3 combined with MMP-9 silencing significantly inhibited the proliferation activity and G1/S cycle progression of the highly metastatic breast cancer cells,and inhibited the expression levels of Ki-67 and CDK1(P<0.05).Furthermore,Rg3 combined with MMP-9 silencing cells inhibited the migration and invasion ability of the cells as well as the expression levels of VEGF and Twist1 proteins(P<0.05).Conclusion The ginsenoside Rg3 can inhibit the invasion and migration of high metastatic breast cancer cells by inhibiting MMP-9 expression.The ginsenoside Rg3 combined with silencing MMP-9 can inhibit the growth,migration and invasion of breast cancer cells.
