摘 要:目的:探讨热性中药导致虚热证及寒性中药导致虚寒证小鼠模型的生物学表达特征。方法:SPF级C57/BL健康小鼠随机分为虚热模型组、虚寒模型组、正常组,虚热组采用灌胃热性中药复方,建立虚热证动物模型,虚寒组采用灌胃寒性中药复方,建立虚寒证动物模型,正常组每天灌以等量的0.9%氯化钠溶液水1次,连续18d,观察并记录小鼠的一般情况和行为;利用放免法检测环核苷酸(cAMP、cGMP),甲状腺素(T4)、皮质酮(CORT),白细胞介素(IL)-6,IL-1β,利用生化法检测ATP酶(ATPase)、琥珀酸脱氢酶(SDH)、乳酸脱氢酶(LDH)、丙酮酸、肝糖原、乳酸。结果:与正常组比较,虚热组小鼠肛温(RTP)、cGMP、IL-6、cAMP、T4、CORT、趾温(TTP)、丙酮酸、LDH、进食量、ATPase、cAMP/cGMP值升高(P<0.01,P<0.05),小鼠去热区的次数与去冷区的差值(CHT)、IL-1β降低(P<0.01,P<0.05);与正常组比较,虚寒组小鼠TTP、cGMP、IL-1β值降低(P<0.01,P<0.05),CHT、肝糖原、乳酸值升高(P<0.01,P<0.05)。结论:小鼠虚热证模型和虚寒证模型造模成功,热性中药致虚热小鼠和寒性中药致虚寒小鼠的各个生理指标有偏离正常水平的趋势。Objective:To discuss biological expression characteristics of mice model of deficiency heat syndrome caused by febrile traditional Chinese medicine and asthenia cold syndrome result from the cold property traditional Chinese medicines.Methods:C57/BL healthy mice with the level of SPF were randomly divided into three groups:normal group,deficiency heat syndrome group,asthenia cold syndrome group.Mice model of deficiency heat syndrome were established by intragastric administration of heat Chinese herbal compound for 18 days.The asthenia cold syndrome group was intragastrically administered with cold Chinese herbal compound for 18 days.In the blank group,mice were given the same amount of physiological saline by gavage once a day for 18 days.The general conditions and the behavior of mice,such as weights,rectal temperature,toe temperature,autonomous activities were observed and recorded.The levels of cGMP,cAMP,Tetraiodothyronine(T4),Corticosterone(CORT),interleukin-6(IL-6),interleukin-1β(IL-1β)were detected by radioimmunoassay,the level of ATPase,succinic dehydrogenase(SDH),Lactate Dehydrogenase(LDH),Pyruvic acid,liver glycogen and lactic acid were detected by biochemistry.Results:Compared with the normal group,the level of anorectal temperature,cGMP,IL-6,cAMP,T4,and CORT,toe temperature,pyruvate,LDH,food intake,ATPase,cAMP/cGMP in deficiency heat syndrome group increased(P<0.01,P<0.05),and the level of the difference between the frequency in the hot area and the frequency in the cold area(CHT),IL-1βdecreased(P<0.01,P<0.05).Compared with the normal group,the level of toe temperature,cGMp,IL-1βin asthenia cold syndrome group decreased(P<0.01,P<0.05),CHT,glycogen,lactate value increased(P<0.01,P<0.05).Conclusion:The mice models of deficiency heat syndrome and asthenia cold syndrome were successfully established and the physiological indexes of the mice deviated from the normal level.
