摘  要:目的:探讨儿茶酚-O-甲基转移酶基因158号密码子(COMT rs4680)基因多态性及血瘀质生物学基础与小鼠肺癌发生发展的关系。方法:将80只C57BL/6小鼠随机分为2组,将其中一组构建血瘀质小鼠模型,模型成功后检测血瘀质小鼠与正常小鼠外周血中COMT rs4680单核苷酸多态性(SNP);在血瘀质模型小鼠及正常小鼠复制Lewis肺癌模型,观察并记录不同小鼠肿瘤直径长至1 cm所用的时间;当小鼠肿瘤直径长至1 cm后,取外周血及肿瘤组织,检测肿瘤组织COMT rs4680 SNP,并比较COMT基因型与1 cm成瘤时间的关系;对比分析血瘀质小鼠、正常小鼠、正常小鼠肿瘤组与血瘀质小鼠肿瘤组外周血中COMT蛋白表达情况。结果:血瘀质小鼠外周血COMT突变基因型G/A与A/A比例明显高于正常小鼠组,差异有统计学意义(P<0.05);血瘀质小鼠肿瘤直径长至1 cm时间短于正常小鼠肿瘤组,差异有统计学意义(P<0.05);A/A基因型小鼠肿瘤长至1 cm的时间明显短于G/G与G/A组小鼠,差异有统计学意义(P<0.05);血瘀质小鼠肿瘤组外周血中COMT蛋白表达明显低于其他组。结论:COMT基因表达的下调以及血瘀质体质可能与肺癌的发生发展有关,且COMT rs4680单核苷酸G到A的突变也促进了肺癌的发生与发展。Objective:To investigate the relationship of gene polymorphism of Catechu-o-methyltransferase codon 158(COMT rs4680) and biological basis of blood stasis constitution in the occurrence and development of lung cancer in mice.Methods: A total of 80 C57 BL/6 mice were randomly divided into two groups. Mice in one group were constructed into models with blood stasis constitution;the single nucleotide polymorphism(SNP) of COMT rs4680 in peripheral blood of these mice with blood stasis constitution and of normal mice was detected after successful modeling. The mice with blood stasis constitution and normal mice were replicated Lewis lung cancer model,and the time taken for diameter of tumor up to 1 cm in different mice was observed and recorded. When the diameter of tumor of mice grew to 1 cm,the peripheral blood and tumor tissue were collected to detect SNP of COMT rs4680, and the relationship between COMT genotype and tumor formation time of 1 cm was compared. The protein expression of COMT in peripheral blood of mice with blood stasis constitution, normal mice, normal mice of tumor group and mice with blood stasis constitution of tumor group was compared and analyzed. Results:The ratio of G/A and A/A of COMT mutant genotype in peripheral blood of mice with blood stasis constitution was significantly higher than that of normal mice,the difference being significant(P<0.05). The diameter of tumor of mice with blood stasis constitution was up to 1 cm,longer than that of normal mice of tumor group,the difference being significant(P<0.05). The time taken for diameter of tumor up to 1 cm in A/A genotype group was significantly shorter than that in G/G and G/A groups, the difference being significant(P<0.05). The protein expression of COMT in peripheral blood of mice with blood stasis constitution of tumor group was significantly lower than those in other groups.
