摘  要:【目的】InDel(insertion/deletion,插入/缺失)在基因组上数量多、分布广。InDel标记是近年来发展的一种新型共显性分子标记,以PCR技术为基础,检测方便、经济有效,且准确性高、稳定性好。因此,InDel标记已经广泛应用于动植物的基因型鉴定、遗传多样性分析、基因定位以及功能标记开发等。但是,目前菜豆资源遗传变异的InDel分子标记研究尚知之甚少。【方法】利用已报道的菜豆InDel序列,合成InDel引物,通过普通PCR扩增,检测25份菜豆种质资源的InDel多态性位点,分析遗传多样性参数,进行UPGMA聚类分析,结合菜豆籽粒农艺性状评价,研究菜豆种子遗传变异特点。【结果】25份菜豆的种子表型差异明显,包括单粒长度、单粒宽度、单粒厚度、百粒质量等。种皮的颜色大致可划分为纯色和花色两个系列。纯色主要包括为黄色(7份)、黑色(5份)、白色(4份)和红色(1份);种皮花色的类型包括红色带黄斑(4份)、黄色带红斑(2份)、红色带白斑(1份)和黄色带白斑(1份)。12对InDel多态性引物在25份菜豆中,共探测到26个InDel位点,平均位点数是2.17个。引物的平均有效等位位点数为1.78个,多态信息含量PIC平均值是0.33,期望杂合度(He)平均值为0.42,观察杂合度(Ho)平均值为0.52,群体的遗传多样性指数I为0.63。UPGMA聚类分析表明,在遗传相似性系数为0.54时,25份菜豆资源可分为5个类群,聚类结果与其种皮颜色存在一定相关性。【结论】12个InDel标记检测出25份供试菜豆资源的中等遗传多样性水平,并揭示其种皮颜色的遗传分化特点。这将有助于菜豆遗传资源的评价、鉴定和保护利用等分子生物学研究。[Objective] InDel(insertion/deletion)exists widely in genome with a large number.InDel is a new type of co-dominant molecular markers developed in recent years.Based on PCR technology,it is easy to bedetected cost-effectively,and shows high accuracy and good stability.Therefore,In Del markers have been wide-ly used in animal and plant studies,including genotyping,genetic diversity analysis,gene mapping,and func-tional marker development.However,knowledge is limited in the current In Del molecular marker studies on thegenetic variation of kidney bean germplasm resources[.Method]Using the published In Del sequences in kid-ney bean,a set of In Del primers were synthesized and amplified by ordinary PCR.25 kidney bean accessionswere analyzed by the In Del primers and their genetic diversity were evaluatedbase on several genetic diversityparameters.Then,UPGMA clustering analysis was carried out to investigate the genetic variation characteristicsof kidney bean seeds,combining with kidney bean grain agronomic traits[.Result]The seed phenotypes of 25 kidney bean accessions showed obviousdifference,including single grain length,single grain width,single grainthickness,and 100 grain weight.The seed coatcolors can be generally divided into two types:singular color andmosaic color.Singular color types mainly included yellow(7 accessions),black(5 accessions),white(4 acces-sions)and red(1 accession);and the types of seed mosaic colorsinclude red with yellow spots(4 accessions),yellow with red spots(2 accessions),red with white spots(1 accession)and yellow with white spots(1 acces-sion).Twelve pair primers detected 26 polymorphic In Del loci in 25 kidney bean accessions,with an average of2.17 loci/In Del.The average number of effective alleles of the primers was 1.78,the average polymorphic infor-mation content PIC was 0.33,the expected average heterozygosity(He)was 0.42,the average observed heterozy-gosity(Ho)was 0.52,and the population genetic diversity index I was 0.63.UPGMA cluster analysis showed thatat the genetic similar
