摘 要:目的探讨恒清方对脑缺血再灌注损伤的保护作用及其机制。方法将48只雄性SD大鼠随机分为假手术组、模型组、恒清方低剂量组、恒清方中剂量组、恒清方高剂量组和尼莫地平组,各8只。造模前1周灌胃给药(低剂量、中剂量、高剂量恒清方或尼莫地平),假手术组与模型组给予等体积生理盐水。末次给药30 min后,建立大鼠大脑中动脉局灶性栓塞(MCAO)模型,造模1.5 h再灌注24 h后,评估大鼠神经功能,测定脑指数、脑含水量、脑梗死面积,采用酶联免疫吸附法测定(ELISA)检测脑组织超氧化物歧化酶(SOD)活性、丙二醛(MDA)、一氧化氮(NO)及血清白介素-6(IL-6)、白介素-1β(IL-1β)、肿瘤坏死因子(TNF)水平。结果恒清方可降低大鼠神经功能评分、脑指数、脑含水量(P<0.05或P<0.01),减小脑梗死面积(P<0.05或P<0.01),提高脑组织SOD活性(P<0.05或P<0.01),降低MDA、NO水平(P<0.05或P<0.01),降低血清IL-6、TNF、IL-1β水平(P<0.05或P<0.01),且量效关系呈正相关,高剂量恒清方作用优于尼莫地平(P<0.05)。结论恒清方可改善大鼠脑缺血再灌注损伤,且量效关系呈正相关,其机制可能与抑制炎症反应和抗氧化作用有关。Objective To investigate the effects and mechanism of Hengqing formula on cerebral ischemia-reperfusion injury.Methods Forty-eight male SD rats were randomly divided into the model group,sham operation group,nimodipine group,Hengqing formula group(low,medium and high dose),8 rats in each group.They were orally administered with low,medium or high dose Hengqing formula or nimodipine daily for 1 week,the model group and sham operation group were given the same volume of normal saline before they were made by middle cerebral artery occlusion(MCAO).After the last administration for 30 minutes,the MCAO model was established,and models were constructed for 1.5 h and reperfused for 24 h.The neurological function score,brain index,brain water content and cerebral infarct volume were measured.Superoxide dismutase(SOD),malondialdehyde(MDA),nitric oxide(NO),interleukin-1β(IL-1β),tumor necrosis factor(TNF),and interleukin-6(IL-6)were detected by Enzyme-linked immunosorbent assay.Results Hengqing formula improved the neurological function score,decreased the brain index,brain water content(P<0.05 or P<0.01),reduced the cerebral infarct volume(P<0.05 or P<0.01),increased the activity of SOD in brain tissue(P<0.05 or P<0.01),reduced the levels of MDA and NO in brain tissue(P<0.05 or P<0.01),and reduced the levels of IL-6,TNF,and IL-1β(P<0.05 or P<0.01).Conclusion Hengqing formula could improve cerebral ischemia-reperfusion injury in rats,which presented positive correlation in dose-effect relationship,and the mechanism might be related to the inhibition of inflammatory response and antioxidant effect.
