摘 要:目的探讨白藜芦醇对马凡氏综合征(MFS)动脉瘤凋亡作用机制的影响。方法体外培养原代血管平滑肌(VSMCs)细胞,实验分为4组:Control组,培养野生小鼠原代VSMCs细胞;Control+Resveratrol组,基于Control组加入白藜芦醇处理细胞;MFS组,培养马凡小鼠原代VSMCs;MFS+Resveratrol组,基于MFS组加入白藜芦醇处理细胞。各组处理24 h后,采用MTT检测细胞增殖率;采用流式细胞术检测细胞凋亡率;采用Western blot检测细胞的凋亡蛋白Bax、Bcl-2的表达。结果MTT结果显示:与Control组比较,MFS组的细胞增殖率明显上升(P<0.05);与MFS组比较,MFS+Resveratrol组的细胞增殖率明显下降(P<0.05)。流式细胞术结果显示:与Control组比较,MFS组的凋亡率明显下降(0.0604±0.0067与0.0243±0.0068,P<0.001);与MFS组相比,MFS+Resveratrol组的凋亡率明显上升(0.0243±0.0068与0.1201±0.0163,P<0.001)。Western blot结果显示:与Control组比较,MFS组的Bax/Bcl-2明显下降(P<0.05);与MFS组相比,MFS+Resveratrol组的Bax/Bcl-2明显上升(P<0.05)。结论白藜芦醇通过上调Bax蛋白的表达进而促进血管平滑肌细胞凋亡反应,最终改善MFS动脉瘤形成过程。Objective To the effect of resveratrol on the apoptosis mechanism of Marfan syndrome(MFS)aneurysm.Methods Primary vascular smooth muscle(VSMCs)cells were cultured in vitro.The experiment was divided into 4 groups.Primary VSMCs cells of wild mice were cultivated in control group.The cells were treated with resveratrol based on the control group in control+resveratrol group.Primary VSMCs of Marfan mice were cultured in MFS group.The cells were treated with resveratrol based on MFS group in MFS+resveratrol group.VSMCs proliferative rate was detected by the MTT assays.Apoptosis level was detected by flow cytometry after treatment for 24 h in each group.The ratio of apoptosis proteins Bax to Bcl-2 was detected by western blot.Results MTT results showed that the proliferative rate of VSMCs in the MFS group was significantly higher than that in the control group(P<0.05).Compared with the MFS group,the proliferative rate in the MFS+resveratrol group was significantly decreased(P<0.05).Flow cytometry results showed that the apoptotic rate of the MFS group was significantly lower than that of the control group(0.0604±0.0067 vs 0.0243±0.0068,P<0.001).Compared with the MFS group,the apoptosis rate of the MFS+resveratrol group was significantly increased(0.0243±0.0068 vs 0.1201±0.0163,P<0.001).Western blot results showed that Bax/Bcl-2 was significantly decreased in the MFS group compared with the control group(P<0.05).Compared with MFS group,Bax/Bcl-2 in the MFS+resveratrol group was significantly increased(P<0.05).Conclusion Resveratrol could promote the apoptosis of vascular smooth muscle cells by up-regulating the expressions of Bax protein,which might eventually improve the formation of Marfan syndrome aneurysms.
