摘  要:以分离鉴定出的核桃黑斑病病原菌——黄单胞杆菌(Xanthomonas campestris)为试验菌株,制备该致病细菌的胞外多糖溶液,探究其致病性并测定其对核桃叶片相关生理指标的影响。结果表明:黄单胞杆菌胞外多糖溶液能使核桃呈现典型的黑斑症状,显著增加细胞膜通透性和丙二醛质量摩尔浓度。在一定质量浓度范围内,核桃黄单胞杆菌胞外多糖与核桃叶片游离脯氨酸质量分数和防御酶活性呈正相关;而当质量浓度较高时,胞外多糖与游离脯氨酸质量分数以及防御酶活性呈负相关,这说明高质量浓度的胞外多糖溶液能影响核桃机体的正常生理指标,从而证明黄单胞杆菌胞外多糖在其对核桃致病过程中具有重要作用。In order to investigate the pathogenic role of the pathogenic bacteria extracellular polysaccharides in walnut black spot,with walnut we isolated and identified the black spot pathogen of the walnut black spot Xanthomonas as a test strain to prepare the pathogenic bacteria extracellular polysaccharide solution,explored its pathogenicity and determined its effect on related physiological indexes of walnut leaves.The results showed that X.exopolysaccharide solution can make walnuts show typical symptoms of dark spots,and significantly increase cell membrane permeability and malondialdehyde content.Within a certain concentration range,the extracellular polysaccharides of X.solani were positively related to the free proline content and defense enzyme activity of walnut leaves;while at higher concentrations,the extracellular polysaccharide was related to free proline content and defense enzyme activity.Negative correlation,which indicates that the high concentration of extracellular polysaccharide solution can affect the normal physiological indicators of walnut,and proves that the extracellular polysaccharide of pathogenic bacteria has an important role in the pathogenic process of walnut by Xanthomonas.
