摘 要:目的探讨非小细胞肺癌(NSCLC)患者血清外泌体中表皮生长因子受体(EGFR)的表达情况以及对NSCLC细胞NCI-H1299增殖和侵袭的影响。方法选取10例原发NSCLC患者(NSCLC组)及8例健康者(正常组)的血清标本,提取分离血清中的外泌体并进行鉴定后,检测外泌体中EGFR蛋白的表达水平。收集15例NSCLC患者的癌组织和癌旁组织,检测组织中EGFR mRNA的表达水平,并分析NSCLC患者血清外泌体EGFR蛋白表达水平与NSCLC组织EGFR mRNA表达水平的相关性。取对数生长期NCI-H1299细胞,分为NSCLC组、正常组、对照组,NSCLC组、正常组细胞分别加入NSCLC患者、正常健康人血清外泌体悬液,检测培养24 h、48 h、72 h、96 h后细胞增殖情况,以及培养48 h后细胞的侵袭能力。结果NSCLC组外泌体中EGFR蛋白的相对表达水平高于正常组,NSCLC组织中EGFR mRNA相对表达水平高于癌旁组织,两者呈正相关(均P<0.05)。NSCLC组NCI-H1299细胞培养48 h、72 h、96 h后的细胞增殖情况以及培养48 h后的侵袭能力均高于其他两组(均P<0.05)。结论NSCLC患者血清外泌体中EGFR蛋白表达上调,且与肿瘤组织中的EGFR mRNA表达呈正相关;NSCLC患者血清外泌体可促进NCI-H1299细胞增殖和侵袭能力。Objective To investigate the expression of serum exosome epidermal growth factor receptor(EGFR)and its effect on the proliferation and invasion of NCI-H1299 cells in patients with non-small cell lung cancer(NSCLC).Methods Serum samples were collected from 10 patients with primary NSCLC(NSCLC group)and 8 healthy individuals(normal group)to perform exosome extraction and isolation derived from serum prior to identification,the expression level of EGFR protein in exosome was measured.Cancer tissue and paracancerous tissue were collected from 15 NSCLC patients to detect the expression levels of their EGFR mRNAs,and the correlation of serum exosome EGFR protein expression level with NSCLC tissue EGFR mRNA expression level was analyzed in NSCLC patients.The NCI-H1299 cells at logarithmic growth phase enrolled were divided into NSCLC group,normal group and control group,cells in the NSCLC and normal groups were added with serum exosome suspensions derived from NSCLC patients and normal healthy individuals,respectively,cell proliferation 24,48 and 96 hours after culture,as well as cell invasion ability 48 hours after culture,was detected.Results The relative expression level of exosome EGFR protein in the NSCLC group was higher than that in the normal group,the relative expression level of EGFR mRNA in NSCLC tissue was higher than that in paracancerous tissue,and a positive correlation was observed between the two levels(all P<0.05).The NSCLC group exhibited increased cell proliferation 48,72 and 96 hours after NCI-H1299 cell culture,as well as increased invasion ability 48 hours after culture,as compared with the other two groups(all P<0.05).Conclusion Serum exosome EGFR protein expression is upregulated in NSCLC patients,which positively correlates with tumor tissue EGFR mRNA expression.Serum exosome might promote the proliferation and invasion ability of NCI-H1299 cells in NSCLC patients.
