摘　　要：目的探讨miR-451a和MMP-2蛋白在结直肠癌组织中的表达情况及两者对肿瘤细胞增殖的影响。方法采用逆转录聚合酶链反应(RT-PCR)和免疫印迹分别检测结直肠癌组织与癌旁组织(来自2018年金华市中心医院手术切除的标本),多个肿瘤细胞系中miR-451a的表达和MMP-2蛋白的表达;在HCT116细胞系中过表达miR-451a(miR-451a-mimic)检测MMP-2蛋白表达;miR-451a-mimic或沉默MMP-2表达检测细胞增殖情况。结果在结直肠癌组织中miR-451a表达明显下降,MMP-2蛋白的表达显著升高,这种结果同时出现在各个结直肠癌细胞株中,以HCT116细胞株最为明显;在HCT116细胞系中miR-451a-mimic显著减少MMP-2蛋白表达量,miR-451a-mimic或沉默MMP-2均会抑制肿瘤细胞的增殖。结论 miR-451a在结直肠癌组织中表达明显降低,其本身可能通过抑制MMP-2蛋白的表达来抑制肿瘤细胞的增殖。Objective To investigate the expression of miR-451 a and MMP-2 proteins in colorectal cancer tissues and their effects on tumor cell proliferation. Methods Reverse transcriptase polymerase chain reaction(RT-PCR) and im munoblotting were used to detect the expression of miR-451 a and MMP-2 proteins in multiple tumor cell lines in col orectal cancer tissues and paracancerous tissues(from the specimens surgically resected by Jinhua Central Hospital in2018), respectively. The expression of MMP-2 protein was detected by over-expression of miR-451 a(miR-451 amimic) in HCT116 cell lines;cell proliferation was detected by miR-451 a-mimic or by silencing MMP-2 expression.Results The expression of miR-451 a in colorectal cancer tissues significantly decreased, while the expression of MMP-2 protein significantly increased;this result was found in all colorectal cancer cell lines simultaneously, with the HCT116 cell line being the most obvious;miR-451 a-mimic significantly reduced the expression of MMP-2 protein in HCT116 cell lines, and miR-451 a-mimic or silencing MMP-2 inhibited the proliferation of tumor cells. Conclusion The expression of miR-451 a in colorectal cancer tissues is significantly reduced, which itself may inhibit the proliferation of tumor cells by inhibiting the expression of MMP-2 protein.
