摘  要:目的NT-3联合NSCs较单纯NSC移植能促进NSCs定向分化成OLs,促进缺血缺氧鼠的神经功能恢复。方法进行鼠胎脑皮层组织NSCs的体外培养、传代,然后将培养好的NSCs制成单细胞悬液,出生7 d的Wistar大鼠60只随机数字表法分成4组。采用经典模型方法制备缺血缺氧模型,正常组老鼠不给予任何干预措施,鉴定模型成功后行立体定向移植,分别移植到鼠侧脑室内生理盐水、NSCs、NT-3和NSCs混合液。移植后4周断头取脑免疫组化染色病理比较。结果免疫组化模型组较正常组AS阳性数目增加,OLs阳性细胞数减少。移植组较模型组AS有所减少,OLs较模型组增多。NT-3联合组恢复更好。结论NT-3能够通过促进NSCs的增殖和诱导分化作用,促进NSCs向OLs分化,对缺血缺氧脑损伤患者的神经功能恢复起重要作用,为今后治疗HIE提供了理论依据。Objective This work was undertaken to determine the effect of neurotrophin-3 combined with neural stem cell can promote NSCs directional differentiation into oligodendrocytes and promote in recovery of neurological function of hypoxic-ischemic brain injury in new born rat.Methods Rat fetal cerebral cortical tissue NSCs cultured in vitro and passage,then cultured NSCs into single cell suspension.A total of 60 postnatal 7 days using a random number table will be divided four groups.The classical model method of oxygen preparation ischemia model,the normal mice don't give in any intervention measures,appraisal model success before the stereospecific transplantation,were transplanted into rat lateral intraventricular physiological saline,NSCs transplantation,NT-3 and NSCs mixture.4 weeks after transplantation,the brains were removed for gross specimens,immunohistochemical staining pathological comparison.Results Immune groupsThe model group MBP positive neurocytes in the periventricular zone on 3-week were fewer,to compared with the normal group on 3-week.For NSCs transplant group the MBP positive cell is more than B group but fewer than normal group.It is the same with D group,and the positive cell is more than NSC transplant group,but GFAP positive neurocytes on the contrary.Conclusion NT-3 promoted NSCs proliferation and differentiation-inducing effect can further promote the recovery of neurological function of hypoxic-ischemic brain injury patients,and provide an effective approach for the treatment of cerebral palsy.
