摘  要:试验建立了气相色谱(GC)法测定动植物中角鲨烯含量的方法。非油脂类试样经水解,乙醚-石油醚混合溶液提取其中的脂肪后,在碱性条件下皂化和甲酯化;不可皂化物角鲨烯用正己烷萃取,用气相色谱法进行测定;动植物油脂试样不经脂肪提取,直接皂化和甲酯化,后同非油脂类样品分析过程。在此试验条件下,角鲨烯检出限(S/N=3)为0.5 mg/100 g,定量限(S/N=10)为1.5 mg/100 g,其质量浓度在3.00~100μg/mL范围内呈现良好线性关系,相关系数r=0.9999,该方法精密度试验结果相对标准偏差(RSD)为1.30%(N=6),在加标浓度为1.30~447 mg/100 g条件下,方法回收率在91.5%~99.7%之间,RSD≤3.26%(N=6)。该方法处理样品基质干扰小,灵敏度高,避免检测器污染,可广泛地应用于动植物中角鲨烯含量的检测。A gas chromatography method is established to determine the contents of squalene in plants and animals.The non-lipid samples are acid-hydrolyzed,the ether is extracted from the ether-petroleum ethersolution,then saponified and methylated under alkaline conditions,and the saponified squalene was extracted with n-hexane and determined by gas chromatography;The animal and vegetable oil samples are directly saponified and methylated without fat extraction,then the analysis process is the same as that of non-grease samples.Under the experimental condition,the limit of detection(S/N=3)was 0.5 mg/100 g,the limit of quantitation(S/N=10)was 1.5 mg/100 g,and it showed a good linear relationship in the range of 3.00-100μg/mL(r=0.9999).The RSD of the accuracy test result was 1.30%.The recovery was between 91.5%and 99.7%%,and the RSD was less than 3.26%in the added concentration range of 1.30-447 mg/100 g(N=6).The method had the advantage in weak matrix interference,high sensitivity,and avoiding contamination detector,and it could be widely used in the analysis of squalene in plants and animals.
