摘 要:目的探讨低温等离子体对人骨肉瘤细胞株MG63的杀伤作用及其可能作用机制。方法采用MTT法和克隆形成实验测定低温等离子体对人骨肉瘤细胞株MG63的增殖抑制作用,显微镜下观察低温等离子体对MG63细胞形态的影响,流式细胞术检测细胞凋亡,Western bolt检测凋亡相关蛋白的表达变化。结果显微镜观察结果显示低温等离子体可以诱导骨肉瘤MG63细胞发生皱缩,在MTT和克隆形成实验中都显示出低温等离子体对骨肉瘤细胞MG63具有明显的增殖抑制效果并且呈剂量依赖性,流式细胞分析显示低温等离子体是通过诱导细胞凋亡而引起细胞增殖抑制。Western blot法显示低温等离子体可以诱导凋亡蛋白的表达,并且呈剂量依赖性。结论低温等离子体对MG63细胞株具有显著的杀伤作用,可能与诱导细胞凋亡有关。Objective To investigate the killing effect of Cold atmoshperic plasma on human osteosarcoma cell line MG63 and its possible mechanism. Methods The proliferation inhibition effect of Cold atmoshperic plasma on MG63 cells was determined by MTT assay and colony formation assay. The effect of Cold atmoshperic plasma on the morphology of MG63 cells was observed under a microscope. Apoptosis was detected by flow cytometry, and the changes in expression of apoptosis related proteins was detected by Western blot. Results Microscopic observation showed that Cold atmoshperic plasma could induce the shrinkage of MG63 cells. In both MTT and colony formation experiments, Cold atmoshperic plasma showed significant proliferation inhibition effect on MG63 cells in a dose-dependent manner. The cellular analysis showed that Cold atmoshperic plasma caused inhibition of cell proliferation by inducing apoptosis. Western blot showed that Cold atmoshperic plasma could induce the expression of apoptotic proteins in a dose-dependent manner. Conclusion Cold atmoshperic plasma has a significant killing effect on MG63 cells, which may be related to the induction of apoptosis.
