摘 要:目的探讨电针对创伤后应激障碍(PTSD)样大鼠学习记忆及海马CA1区Bcl-2/Bax表达的影响。方法将48只健康清洁级雄性SD大鼠随机均分为4组:正常对照组、模型组、电针组和电针对照组,采用国际认定的单次延长应激(SPS)方法制作大鼠PTSD模型,在造模6 h后给予电针组和电针对照组针刺治疗,通过旷场实验和Morris水迷宫实验测试大鼠的情绪唤醒水平和学习记忆能力,采用HE染色观察大鼠海马组织CA1区组织形态变化,Western blot实验检测大鼠海马组织CA1区Bcl-2/Bax蛋白的表达。结果与模型组大鼠比较,正常对照组和电针组大鼠站立与跨格次数均增多,平均上台潜伏期和穿台潜伏期缩短,穿台次数增加,差异均有统计学意义(P<0.05);HE染色结果表明,与正常对照组比较,模型组大鼠海马组织CA1区海马经元排列紊乱,结构异常,电针组CA1区海马神经元排列较整齐,结构趋于正常;与正常对照组比较,模型组大鼠海马组织CA1区Bcl-2/Bax蛋白表达升高,与模型组比较,电针组大鼠海马组织CA1区Bcl-2/Bax蛋白表达降低,差异有统计学意义(P<0.05)。结论电针治疗可改善PTSD样大鼠的学习记忆能力,可能与下调海马组织CA1区Bcl-2/Bax蛋白表达有关。Objective To investigate the effects of electro-acupuncture on learning and memory and Bcl-2/Bax expression in hippocampal CA1 region of post-traumatic stress disorder(PTSD)-like rats.Methods Forty-eight healthy and clean male SD rats were randomly divided into four groups:control group,model group,electro-acupuncture group and electro-acupuncture control group,12 rats in each group.The PTSD model of rats was established by the internationally recognized single prolonged stress(SPS)method.After 6 h of modeling,the rats in the electro-acupuncture group and electro-acupuncture control group were given acupuncture treatment.The open field test and Morris water maze were used to test the emotional arousal level and learning and memory ability of rats.The morphological changes of hippocampal CA1 region were observed by HE staining.Then the expression of Bcl-2/Bax protein in hippocampal CA1 region was detected by Western blot.Results Compared with the model group,the standing and crossing times of the control group and the electro-acupuncture group increased,the average incubation period and crossing latency shortened,the number of crossing times increased,and the difference was statistically significant(P<0.05).The results of HE staining showed that compared with the control group,the hippocampal neurons in CA1 area of the model group were disorderly arranged and had an unusual structure,and the hippocampal neurons in CA1 area of the electro-acupuncture group were arranged in order and the structure tended to be normal.Compared with the control group,the expression of Bcl-2/Bax protein in CA1 region of hippocampus in the model group increased,and the expression of Bcl-2/Bax protein in CA1 region of hippocampus in the electro-acupuncture group decreased compared with the model group,and the difference was statistically significant(P<0.05).Conclusion Electro-acupuncture can improve the learning and memory ability of PTSD-like rats,which may be related to the down-regulation of Bcl-2/Bax protein expression in CA1 region
