摘  要:目的观察癫痫大鼠皮层及海马神经细胞内游离Ca^2+水平及低剂量伽玛刀照射对其的影响。方法选取Wistar大鼠32只,随机均分为4组:正常对照组、伽玛刀对照组、戊四氮(PTZ)致痫组、PTZ致痫+伽玛刀组。采用连续腹腔注射PTZ溶液制备癫痫大鼠模型,应用Leksell伽玛刀系统对大鼠进行低剂量伽玛刀照射12周,照射靶区为大鼠额叶皮层,后断头留取额叶及海马组织,制备神经元细胞悬液,应用共聚焦显微镜检测神经细胞内的游离Ca^2+水平。结果与正常对照组相比,PTZ致痫组额叶及海马神经细胞内的Ca^2+荧光强度升高,差异有统计学意义(P<0.05);与PTZ致痫组相比,PTZ致痫+伽玛刀组神经细胞内的Ca^2+荧光强度降低,差异有统计学意义(P<0.05),伽玛刀对照组与正常对照组相比,神经细胞内的Ca^2+轻度升高,但差异无统计学意义(P>0.05)。结论癫痫大鼠额叶皮层及海马神经细胞内Ca^2+水平升高可能是癫痫发病的重要机制之一;低剂量伽玛刀照射可能通过调节神经细胞内钙稳态而发挥抗癫痫和改善认知的作用。Objective To observe the level of intracellular free Ca^2+ in cortex and hippocampus of epileptic rats and the effect of low dose gamma knife irradiation on it.Methods 32 Wistar rats were randomly divided into four groups:①Normal control group;②gamma knife(GK)control group;③pentylenetetrazole(PTZ)group;④PTZ+GK group.Epileptic rat model was prepared by continuous intraperitoneal injection of PTZ.Leksell gamma knife system was used to irradiate rats with low dose gamma knife for 12 weeks.The target area was bilateral frontal lobe.On the 12th week after gamma knife irradiation,the brains of rats were decapitated and neurons of frontal lobe and hippocampus were acutely isolated.The neuron cell suspension was prepared,and then laser scanning confocal microscopy(LSCM)was used to detect the level of free Ca^2+ in nerve cells.Results Compared with the normal control group,the fluorescence intensity of Ca^2+ in cortex and hippocampal neurons of PTZ group increased significantly(P<0.05).Compared with PTZ group,the fluorescence intensity of Ca^2+ in neurons of PTZ+GK group decreased markedly(P<0.05).The fluorescence intensity of Ca^2+ in neurons of GS control group was slightly higher than that of normal control group,but the difference was not statistically significant(P>0.05).Conclusion The increased Ca^2+ level of neurons in frontal cortex and hippocampus may be one of the important mechanisms of epilepsy.Low dose of gamma knife irradiation may play a role of anti-epileptic and improve cognitive function by affecting intracellular calcium homeostasis.
