摘  要:目的探讨白藜芦醇(Rev)通过调控IL-6/JAK2/STAT3信号通路对卵巢癌SKOV-3细胞增殖迁移的影响。方法根据药物处理将细胞分为SKOV-3空白组(CON)、白藜芦醇低浓度处理组(Rev-L)、白藜芦醇中浓度处理组(Rev-M)和白藜芦醇高浓度处理组(Rev-H)。CCK-8检测SKOV-3细胞的增殖;流式细胞术检测SKOV-3细胞的凋亡;细胞划痕愈合实验检测SKOV-3细胞的迁移;Transwell实验检测SKOV-3细胞的侵袭;Western blot法检测各组细胞的转移相关蛋白、凋亡相关蛋白及IL-6/JAK2/STAT3信号通路相关蛋白的表达水平。结果与CON组比较,白藜芦醇处理组SKOV-3细胞凋亡率增加,增殖能力降低,细胞的迁移侵袭能力减弱;凋亡相关蛋白B淋巴细胞瘤-2(Bcl-2)表达水平下降;Bcl-2家族促凋亡蛋白(Bax)、活化的半胱氨酸的天冬氨酸蛋白水解酶3(cleaved Caspase-3)表达水平上调,转移相关蛋白钙黏着蛋白E(E-cadherin)表达水平上调,N-cadherin和波形蛋白(Vimentin)表达水平下调;IL-6/JAK2/STAT3信号通路活性降低。结论Rev能通过下调IL-6/JAK2/STAT3信号通路活性,促进卵巢癌SKOV-3细胞凋亡,抑制卵巢癌SKOV-3细胞的增殖迁移以及侵袭。Objective To investigate the effect of resveratrol(Rev)on proliferation and migration of ovarian cancer SKOV-3 cells by regulating IL-6/JAK2/STAT3 signaling pathway.Methods According to drug treatment,the cells were divided into SKOV-3 blank group(CON),Rev low concentration group(Rev-L),Rev medium concentration group(Rev-M)and Rev high concentration group(Rev-H).CCK-8 assay was used to detect the proliferation of SKOV-3 cells.Flow cytometry was used to detect the apoptosis of SKOV-3 cells.Healing assay was used to detect the migration of SKOV-3 cells.Transwell test was used to detect the invasion of SKOV-3 cells.Western blot assay was used to detect the expression levels of metastasis-related proteins,apoptosis-related proteins and IL-6/JAK2/STAT3 signaling pathway proteins in the each group.Results Compared with CON group,the apoptosis of SKOV-3 cells increased after Rev treatment,the proliferation,migration and invasion ability were inhibited;B lymphoma-2(Bcl-2)expression level was decreased;Bcl-2 family apoptotic proteins(Bax)and activated Caspase-3(cleaved Caspase-3)expression levels were increased;transfer related protein Ca2+dependent cell adhesion moleculue family-E(E-cadherin)expression level was increased,Ca2+dependent cell adhesion moleculue family-N(N-cadherin)and Vimentin expression levels were increased;and the activity of IL-6/JAK2/STAT3 signaling pathway decreased.Conclusion Rev can down-regulate the activity of IL-6/JAK2/STAT3 signaling pathway,promote the apoptosis of SKOV-3 cells,and inhibit the proliferation,migration and invasion of SKOV-3 cells.
