摘 要:目的观察温度敏感型富血小板血浆凝胶(temperature sensitive platelet-rich plasma,TSPRP)对兔角膜碱烧伤组织血管内皮细胞形成的影响。方法制作兔角膜碱烧伤模型,抽取自体血,离心获得自体富血小板血浆(platelet-rich plasma,PRP),以泊洛沙姆为基质制成TSPRP。实验兔随机分成4组,分别给予生理盐水(NS)、凝胶空白基质(GBM)、PRP和TSPRP滴眼。14 d后观察角膜炎症和溃疡情况,行免疫组织化学染色,计算CD34阳性血管数。结果14 d后,NS与GBM组角膜炎症和溃疡评分以及微血管数和微血管面密度差异均无统计学意义(P>0.05);PRP组和TSPRP组与NS组相比,角膜炎症和溃疡评分以及微血管数和微血管面密度差异有统计学意义(P<0.05);TSPRP组与PRP组比较角膜炎症和溃疡评分以及微血管数和微血管面密度差异有统计学意义(P<0.05)。结论TSPRP能够控制角膜炎症和血管内皮细胞的形成,减少新生血管,效果优于PRP。Objective To observe the effect of temperature sensitive platelet-rich plasma(TSPRP)on theformation of vascular endothelial cells in alkali burned cornea in rabbits.Methods The alkaline burn model ofrabbit cornea was made,autologous blood was extracted,and autologous platelet-rich plasma(PRP)was obtainedby centrifugation. TSPRP was prepared using poloxam as matrix. The experimental rabbits were divided into 4 groups randomly,and were given normal saline(NS),gel blank matrix(GBM),PRP and TSPRP. After 14 days,corneal inflammation and corneal ulcer were observed,immuno-histochemical staining was performed,and thenumber of CD34 positive blood vessels was calculated.Results After 14 days,there was no significant differencein corneal inflammation and ulcer score,microvascular number and surface density between NS and GBM(P >0.05). Compared with NS,there were statistically significant differences in corneal inflammation and ulcer score,microvascular number and surface density in the PRP and TSPRP(P < 0.05). There were statistically significantdifferences in corneal inflammation and ulcer score,microvascular number and surface density between the TSPRPand PRP(P < 0.05).Conclusion TSPRP could control corneal inflammation and the formation of endothelialcell,reduce neovascularization,and is better than PRP.
