慢性牙周炎患者β-catenin,MMP-8表达情况及与病情严重程度的相关性分析

摘　　要：目的:探讨慢性牙周炎患者β-catenin,MMP-8表达情况及与病情严重程度的相关性。方法:选取慢性牙周炎患者50例为研究对象,依据病情程度分为中度牙周炎(MCP,n=23)和重度牙周炎(SCP,n=27),另选健康志愿者25例为对照组。采用酶联免疫吸附法测定各组龈沟液中MMP-8水平、RT-qPCR法检测各组龈沟液和牙龈组织中β-catenin和MMP-8 mRNA水平、Western blotting法检测各组牙龈组织中β-catenin和MMP-8蛋白表达水平、免疫共沉淀法测定SCP组牙龈组织中β-catenin和MMP-8的相互作用。结果:MCP和SCP组龈沟液中MMP-8水平明显高于对照组(P<0.05),且SCP组MMP-8水平明显高于MCP组(P<0.05),龈沟液中MMP-8水平与CAL呈正相关;MCP和SCP组龈沟液和牙龈组织中β-catenin和MMP-8 mRNA水平明显高于对照组(P<0.05),且SCP组β-catenin和MMP-8 mRNA水平明显高于MCP组(P<0.05),β-catenin和MMP-8 mRNA水平与CAL呈正相关;MCP和SCP组牙龈组织中β-catenin和MMP-8蛋白水平明显高于对照组(P<0.05),且SCP组β-catenin和MMP-8蛋白水平明显高于MCP组(P<0.05),牙龈组织中β-catenin和MMP-8蛋白水平与CAL呈正相关;龈沟液和牙龈组织中β-catenin和MMP-8表达水平呈正相关。结论:牙周炎患者龈沟液和牙龈组织中β-catenin和MMP-8的表达水平可反映慢性牙周炎的严重程度,可为牙周炎治疗提供参考。Objective:To investigate the expression ofβ-catenin and MMP-8 in patients with chronic periodontitis and its correlation with the severity of the disease.Methods:50 patients with chronic periodontitis were selected as the study objects,and they were divided into moderate periodontitis(MCP,n=23)and severe periodontitis(SCP,n=27)according to the severity of the disease,and 25 healthy volunteers were selected as the control group.MMP-8 levels in gingival crevicular fluid were determined by ELISA.RT-qPCR was used to detect the levels ofβ-catenin and MMP-8 mRNA in gingival crevicular fluid and gingival tissue in each group.Western blotting was used to detect the expression levels ofβ-catenin and MMP-8 proteins in the gingival tissues of each group.Immunocoprecipitation method was used to determine the interaction betweenβ-catenin and MMP-8 in gingival tissue of SCP group.Results:MMP-8 level in gingival crevicular fluid in MCP and SCP groups was significantly higher than that in the control group(P<0.05).The level of MMP-8 in SCP group was significantly higher than that in MCP group(P<0.05),and the level of MMP-8 in gingival crevicular fluid was positively correlated with CAL.The levels ofβ-catenin and MMP-8 mRNA in gingival crevicular fluid and gingival tissue in the MCP and SCP groups were significantly higher than those in the control group(P<0.05).THE levels ofβ-catenin and MMP-8 mRNA in SCP group were significantly higher than those in MCP group(P<0.05).The levels ofβ-catenin and MMP-8 mRNA were positively correlated with CAL.The levels ofβ-catenin and MMP-8 proteins in the gingival tissues of the MCP and SCP groups were significantly higher than those of the control group(P<0.05).The levels ofβ-catenin and MMP-8 proteins in the gingival tissues of the SCP group were significantly higher than those of the MCP group(P<0.05).CAL was positively correlated with the levels ofβ-catenin and MMP-8 proteins in gingival tissue.There was a positive correlation between the expression levels ofβ-catenin and MMP-8 in
