摘 要:目的探讨没食子酸对放射性肺损伤(RILI)大鼠的保护作用。方法将36只SD雄性大鼠随机分为对照组、RILI组、RILI+没食子酸组,各12只。对RILI组及RILI+没食子酸组,利用单次双肺20 Gy照射制备RILI大鼠模型。从照射后第1天开始,RILI+没食子酸组给予没食子酸灌胃,RILI组给予等量生理盐水灌胃,对照组不做任何处理,灌胃持续至照射后第28天。在照射后第4周末及第8周末,每组各处死6只大鼠,检测血清白细胞介素6(IL-6)、转化生长因子β1(TGF-β1)水平;获取肺组织,检测肺系数及肺组织羟脯氨酸含量、α平滑肌肌动蛋白(α-SMA)表达,观察肺组织病理改变,并行肺泡炎及肺纤维评分。结果在照射后第4周末、第8周末,RILI组及RILI+没食子酸组的肺系数、肺泡炎及肺纤维评分、血清IL-6及TGF-β1水平、肺组织羟脯氨酸含量及α-SMA表达水平均高于对照组,而RILI+没食子酸组以上指标水平均低于RILI组(均P<0.05)。结论没食子酸对RILI大鼠具有保护作用,而这可能是通过减轻炎症、抑制成纤维细胞活化而实现的。Objective To explore the protective effect of gallic acid on rats with radiation-induced lung injury(RILI).Methods Thirty-six SD male rats were randomly divided into control group,RILI group and RILI+gallic acid group,with 12 rats in each group.A rat model of RILI was prepared in the RILI group and the RILI+gallic acid group by single bilateral lung 20 Gy irradiation.From the first day after irradiation,the RILI+gallic acid group received gavage administration of gallic acid,while the RILI group received gavage administration of isometric normal saline,the control group did not receive any treatment,and the gavage administration lasted to the 28th day after irradiation.At the end of the 4th week and the 8th week,six rats were killed in each group,then serum interleukin 6(IL-6)and transforming growth factorβ1(TGF-β1)levels were detected;and lung tissues were obtained,then the lung coefficient,hydroxyproline content andαsmooth muscle actin(α-SMA)expression in the lung tissues were determined,the pathological changes in lung tissues were observed,and scoring for alveolitis and lung fibrosis was performed.Results At the end of the 4th week and the 8th week,the RILI group and the RILI+gallic acid group yielded higher lung coefficient,scores for alveolitis and lung fibrosis,serum IL-6 and TGF-β1 levels,hydroxyproline content andα-SMA expression level in the lung tissues as compared with the control group,and the levels of the indexes mentioned above in the RILI+gallic acid group were lower than those in the RILI group(all P<0.05).Conclusion Gallic acid exerts a protective effect against RILI in rats probably by alleviating inflammation and inhibiting fibroblast activation.
