白藜芦醇治疗重症急性胰腺炎模型大鼠的效果

摘　　要：目的探讨白藜芦醇治疗大鼠重症急性胰腺炎(SAP)的效果及其机制。方法80只SD雄性大鼠随机分为空白对照组、假手术组、模型组和治疗组,每组20只。模型组和治疗组采用逆行胆管注射3%牛黄胆酸钠1 mL/kg建立SAP大鼠模型,治疗组造模成功后12 h,腹腔注射白藜芦醇30 mg/kg治疗。3 d后处死各组大鼠,检测血清脂肪酶和淀粉酶水平,同时对胰腺组织进行HE染色,评估胰腺组织的损伤。采用免疫组化染色观察胰腺组织中血管内皮生长因子A(VEGF-A)、血管内皮生长因子受体2(VEGFR2)以及血管内皮细胞表面分子CD31的表达。另取人脐静脉内皮细胞分为两组行体外培养:给药组加入白藜芦醇(40 mmol/L),对照组不加药物,观察体外血管形成情况。结果与空白对照组和假手术组比较,模型组血清淀粉酶、脂肪酶水平以及组织病理评分升高(P<0.05);治疗组上述指标低于模型组(P<0.05)。免疫组化染色结果显示,与空白对照组和假手术组比较,模型组CD31、VEGF-A和VEGFR2阳性表达减少;治疗组上述指标阳性表达较模型组增多。人脐静脉内皮细胞体外试验显示,给药组人脐静脉细胞血管形成数量多于对照组(P<0.05)。结论白藜芦醇对大鼠SAP有一定的治疗作用,其对胰腺损伤血管的修复可能是其机制之一。Objective To investigate the efficacy of resveratrol in treating rat models with severe acute pancreatitis(SAP).Methods Eighty SD male rats were randomly and equally divided into four groups of A(blank control),B(sham operation),C(model control)and D(resveratrol treatment).The rat models with SAP were established by retrograde injection of 3%sodium taurocholate 1 mL/kg into bile duct in groups of C and D.The rats in group D were injected intraperitoneally with resveratrol 30 mg/kg 12 hours after successful establishment of SAP model.Rats were sacrificed 3 days later.Serum levels of lipase and amylase were detected.The pancreatic tissues were stained with HE and the damages of the pancreas were evaluated pathologically.Immunohistochemical staining was used to observe the expressions of vascular endothelial growth factor A(VEGF-A),vascular endothelial growth factor receptor 2(VEGFR2)and vascular endothelial cell surface molecule CD31 in pancreatic tissues.Human umbilical vein endothelial cells were taken and cultured in vitro,which were divided into two groups of R(resveratrol 40 mmol/L was added in medium)and NR(without resveratrol in medium).The angiogenesis in vitro was observed.Results Compared with groups of A and B,serum levels of amylase,lipase and histopathological scores were increased in group C(P<0.05),which in group D were lower than those in group C(P<0.05).Immunohistochemical staining showed that compared with groups of A and B,the positive expressions of CD31,VEGF-A and VEGFR2 were lower in group C,which were higher in group D than those in group C.Compared with group NR,the number of vascularization of human umbilical vein cells in group R was increased(P<0.05).Conclusion Resveratrol has a certain therapeutic effect on SAP in rats.The one of the mechanisms may be its repair of pancreatic blood vessels.
