摘 要:目的分析多囊卵巢综合征(PCOS)基因转录调控关联的生物信息。方法从整合基因表达汇编平台中共下载PCOS患者33例及健康对照30例的皮下脂肪组织样本转录组测序数据,比较两组基因表达数据,保留P<0.0005的1636个显著差异基因,进行GO数据库的功能富集分析和KEGG数据库的生物信号通路富集分析。结果1636个显著差异基因富集在GO功能条目下,如在生物学过程集合中的嗜同性细胞黏附、肌丝滑动、肌肉收缩等,在细胞组分集合中的细胞质、肌原纤维节、核质等,在分子功能集合中的蛋白质结合、多聚腺嘌呤RNA结合及钙离子结合等(P<0.001)。1636个显著差异基因富集KEGG生物信号通路,如TNF信号通路、催产素信号通路、Ras信号通路、环磷酸腺苷信号通路、胰岛素信号通路及雌激素信号通路等(P<0.05)。结论PCOS表达基因的遗传学变化可能通过影响卵巢发育生物学过程及卵巢发育相关的信号通路的活性,从而促进PCOS的发生。Objective To analyze the biological information related to gene transcription regulation of polycystic ovarian syndrome(PCOS).Methods Transcriptome sequencing data of subcutaneous adipose tissue samples of 33 patients with PCOS and 30 healthy controls were downloaded from the Gene Expression Omnibus platform and the data of gene expressions in both groups were analyzed.A total of 1636 significantly different genes with P<0.0005 was collected.Functional enrichment analysis of GO database and biological signaling pathway enrichment analysis of KEGG database were conducted.Results In functional enrichment analysis of GO database,1636 genes were significantly enriched in term of biological process with homophilic cell adhesion,muscle filament sliding and muscle contraction,in term of cell component with cytosol,sarcomere and nucleoplasm,in term of molecular function with protein binding,poly(A)RNA binding and calciumion binding(P<0.001).In the KEGG-pathway enrichment analysis,1636 genes were significantly enriched in the TNF signaling pathway,oxytocin signaling pathway,Ras signaling pathway,cAMP signaling pathway,insulin signaling pathway and strogen signaling pathway(P<0.05).ConclusionGenetic changes may contribute to the development of PCOS by altering the biological processes of ovarian development and the activity of signaling pathways involved.
