摘 要:目的:研究复方黄芪降糖颗粒(FFHQ)对高血糖模型动物的降血糖作用及其机制。方法:通过尾静脉注射四氧嘧啶致糖尿病小鼠模型和体外HepG2细胞胰岛素抵抗实验,考察空腹血糖、糖耐量等指标,对复方黄芪降糖颗粒降血糖功能性进行评价,同时测定血清胰岛素、肝糖原和肌糖原水平及HepG2细胞胰岛素抵抗情况,对其作用机制进行了初步探讨。结果:复方黄芪降糖颗粒(FFHQ)经给药30天后,复方黄芪降糖颗粒(FFHQ)高、中、低剂量组均能显著降低糖尿病小鼠的空腹血糖、提升糖耐量(p<0.05或0.01),具有较好的降血糖作用。高剂量的FFHQ对正常小鼠的血糖水平无显著的影响。结论:FFHQ具有促进小鼠胰岛素分泌,增加肝糖原和肌糖原含量、改善胰岛素抵抗的作用。Objective:The aim of this study is to investigate the hypoglycemic effect of compound Huangqi hypoglycemic granule (FFHQ) and its mechanism.Method:Alloxan-induced diabetic mice and HepG2 insulin resistance experiments were used to evaluate the functional hypoglycemic effect of FFHQ with the indexes of fasting blood glucose and glucose tolerance etc.The levels of serum insulin,hepatic glycogen and muscle glycogen,and insulin resistance in HepG2 cells were investigated to conduct a preliminary discussion of mechanism of FFHQ.Result:After administration for 30 days,compound FFHQ high,medium and low dose groups can significantly reduce fasting blood glucose and increase glucose tolerance in diabetic mice (p <0.05 or 0.01),which indicated that it had a good hypoglycemic effect.High doses of FFHQ had no significant effect on blood glucose levels in normal mice.Conclusion:FFHQ can promote insulin secretion,increase hepatic glycogen and muscle glycogen content,and improve insulin resistance in mice.
