摘 要:目的:研究蒲公英提取物对阴道加德纳菌(G.vaginalis)的抑制作用及抑制机理。方法:热水浸提蒲公英活性成分,大孔树脂吸附法、萃取法分离纯化蒲公英活性成分,HPLC分析蒲公英活性成分组成;纸片抑菌法研究蒲公英提取物对加德纳菌的抑制作用;分光光度法分析蒲公英提取物的最小抑菌浓度和抑菌动力学曲线;扫描电镜、核酸法和碘化丙啶染色法分析蒲公英提取物对加德纳菌的抑菌机理。结果:以3,5-二-O-咖啡酰奎尼酸为主要成分的蒲公英提取物对加德纳菌有良好的抑制作用,其MIC为32.0 mg·mL-1,抑菌动力学曲线分析表明蒲公英提取物对加德纳菌的抑制作用主要是作用于其指数生长期。扫描电镜和膜通透性分析发现蒲公英提取物可破坏加德纳菌细胞膜结构,增加加德纳菌膜的通透性,进而抑制加德纳菌生长。结论:蒲公英提取物对加德纳菌有明显的抑制效果,该抑制作用是通过破坏菌体细胞膜结构使菌体内容物外泄,进而使细胞生长受到抑制,最终导致其死亡。Objective:To study the antibacterial activity and antibacterial mechanism of dandelion extract against G.vaginalis. Methods:The inhibitory effect of dandelion extract on G.vaginalis was studied by paper bacteriostatic method. On this basis,the minimum inhibitory concentration and antibacterial kinetics of dandelion extract were method. On this basis,the minimum inhibitory concentration and antibacterial kinetics of dandelion extract were analyzed. Scanning electron microscopy analysis,nucleic acid method and propidium iodide staining were used to analyze the antibacterial mechanism of dandelion extract against G.vaginalis. Results:The dandelion extract had a good inhibitory effect on G.vaginalis,and its MIC was 32.0 mg·mL-1. The kinetic curve analysis showed that the inhibitory effect of dandelion extract on G.vaginalis was mainly in its exponential growth phase. Scanning electron microscopy and membrane permeability analysis showed that dandelion extract could destroy the cell membrane structure of G.vaginalis,increase the permeability of G.vaginalis membrane,and then destroy the cell membrane of G.vaginalis to inhibit the growth of G.vaginalis. Conclusion:The dandelion extract has obvious inhibitory effect on G.vaginalis and it can destroy the cell membrane structure and promotes the leakage of cell contents,which inhibits cell growth and eventually leads to death of the cell.
