摘　　要：目的:研究TWIST家族bHLH转录因子1(TWIST family bHLH transcription factor 1,TWIST1)及束蛋白结合蛋白1(Fascin actin-bundling protein 1,FSCN1)mRNA在非小细胞肺癌中的表达及临床意义。方法:利用GEPIA网站分析TWIST1 mRNA和FSCN1 mRNA在非小细胞肺癌中的表达及其表达与患者预后的相关性;利用实时定量PCR检测69对配对肿瘤组织中TWIST1 mRNA及FSCN1 mRNA表达,并结合患者的临床信息分析其表达与患者临床病理特征之间的相关性。结果:GEPIA网站分析显示,TWIST1 mRNA和FSCN1 mRNA在肺腺癌和肺鳞癌组织中表达明显高于癌旁组织(P<0.05)。69对配对肿瘤组织中,非小细胞肺癌组织中TWIST1 mRNA及FSCN1 mRNA表达明显高于对应癌旁组织(P<0.01);二者表达与患者病理分期及有无淋巴结转移相关;二者表达呈正相关(r=0.630,P<0.001)。结论:TWIST1 mRNA及FSCN1 mRNA在非小细胞肺癌中呈高表达且与患者的临床分期及预后相关。Objective:To study the expression and clinical significance of TWIST family bHLH transcription factor 1(TWIST1)and Fascin actin-bundling protein 1(FSCN1)mRNA in non-small cell lung cancer(NSCLC).Methods:The TCGA visualization website GEPIA was used to explore the mRNA expression of TWIST1 and FSCN1 in NSCLC and the correlation between their expression and TNM stages and prognosis of patients;real-time quantitative PCR was preformed to detect the mRNA expression of TWIST1 and FSCN1 in 69 pairs of paired tumor tissues,and the correlation between their expression and the clinicopathological characteristics of patients was analyzed based on the clinical information of patients.Results:The mRNA expression of TWIST1 and FSCN1 in lung adenocarcinoma and lung squamous cell carcinoma was higher than that in adjacent tissues(P<0.05).NSCLC tissues showed higher expression levels of TWIST1 mRNA and FSCN1 mRNA compared with adjacent normal tissues(P<0.05).Both mRNA expression of TWIST1 and FSCN1 were correlated with the pathological stages and lymph node metastasis(P<0.05).Moreover,linear regression analysis showed a significant positive correlation between them(r=0.630,P<0.001).Conclusion:TWIST1 mRNA and FSCN1 mRNA were highly expressed in NSCLC and correlated with the clinical staging and prognosis of patients.
