金丝楠木精油抑制人前列腺癌细胞体外生长的研究

摘　　要：目的:研究金丝楠木精油对人前列腺癌细胞体外生长的影响,为金丝楠木精油预防肿瘤和长寿保健产品的开发与应用提供参考。方法:使用3种不同年份的阴沉金丝楠木、近代金丝楠木和新鲜金丝楠木为实验材料,通过加热升腾的方式提取精油。以人前列腺癌细胞系(PC3、DU145和LNCaP)为检测对象。通过细胞计数试剂盒(cell counting kit-8,CCK-8)法检测金丝楠木精油对前列腺癌细胞的抑制情况,计算半数抑制率(50%inhibition concentration,IC50);使用流式细胞仪检测金丝楠木精油对前列腺癌细胞凋亡的影响,评价金丝楠木精油的抗肿瘤活性。结果:3种金丝楠木精油都可以抑制肿瘤细胞生长,新鲜金丝楠木精油对3种前列腺癌细胞系(PC3、DU145、LNCaP)的IC50分别为0.254200%、0.119600%和0.094270%;近代金丝楠木精油对3种前列腺癌细胞系(PC3、DU145、LNCaP)的IC50分别为0.019960%、0.031750%和0.016550%;阴沉金丝楠木精油对3种前列腺癌细胞系(PC3、DU145、LNCaP)的IC50分别为0.008897%、0.016850%和0.004589%,其中效果最好的是阴沉金丝楠木精油,且万分之一含量的阴沉金丝楠木精油可显著诱导人前列腺癌细胞的凋亡,提示金丝楠木精油可能通过诱导凋亡达到抑癌的作用。结论:金丝楠木精油可抑制前列腺癌细胞增殖,促进癌细胞发生凋亡。Objective:To study the inhibitory effect of Phoebe Zhennan essential oil on prostate cancer,in order to provide reference for the development and utilization of Phoebe Zhennan anti-cancer products.Methods:Ancient buried Phoebe Zhennan,recent Phoebe Zhennan and fresh Phoebe Zhennan were used as materials,and extracted essential oil by the way of heating up.Next,three prostate cancer cell lines(PC3,DU145 and LNCap)were taken as the test objects,and detected the inhibition of the essential oil of Phoebe Zhennan on prostate cancer cell by cell counting kit-8(CCK-8)method.Then,the 50%inhibition concentration(IC50)was calculated.After that,flow cytometry was used to detect the influence of Phoebe Zhennan essential oil on apoptosis in prostate cancer cells and the anti-tumor activity of Phoebe Zhennan essential oil was evaluated.Results:All of three kinds of Phoebe Zhennan essential oil can inhibit the growth of tumor cells.The IC50 of fresh Phoebe Zhennan essential oil for three prostate cancer cell lines(PC3,DU145 and LNCap)are 0.254200%,0.119600%and 0.094270%.The IC50 of recent Phoebe Zhennan essential oil for three prostate cancer cell lines(PC3,DU145 and LNCap)are 0.019960%,0.031750%and 0.016550%.The IC50 of ancient buried Phoebe Zhennan essential oil for three prostate cancer cell lines(PC3,DU145 and LNCap)are 0.008897%,0.016850%and 0.004589%.Among these kinds of essential oil,ancient buried Phoebe Zhennan has the best effect,and one in ten thousand of it can significantly promote the apoptosis in prostate cancer cells.Then ancient buried Phoebe Zhennan may achieve the function of anti-cancer by promoting the apoptosis.Conclusion:Phoebe Zhennan essential oil can inhibit the proliferation and promote the apoptosis of prostate cancer cells.
