摘 要:目的评估宫颈癌(CC)患者癌组织长链非编码RNA牛磺酸上调基因1(lncRNA TUG1)的表达水平与CC诊断及预后的关系。方法前瞻性队列研究连续纳入137例接受手术治疗的CC患者。记录所有患者的人口学和临床病理特征并计算患者总体生存期(OS)。采用定量聚合酶链式反应(qPCR)检测CC患者癌组织和癌旁组织lncRNA TUG1的相对表达量。结果CC癌组织的lncRNA TUG1相对表达量显著高于配对的癌旁组织(P<0.001)。癌组织lncRNA TUG1表达水平与国际妇产科联盟(FIGO)分期(P=0.023)呈正相关。Kaplan-Meier(K-M)曲线显示,癌组织lncRNA TUG1高表达的患者OS显著低于癌组织lncRNA TUG1低表达的患者(P<0.001)。多元Cox比例风险模型分析显示,癌组织lncRNA TUG1高表达是CC患者OS较差的独立预测因素(P<0.001)。肿瘤大小≥4 cm(P<0.001)、FIGOⅢ期(P<0.001)、淋巴结转移阳性(P<0.001)和宫颈基层浸润深度≥2/3(P=0.002)也是CC患者OS较差的独立预测因素。结论癌组织lncRNA TUG1表达水平可作为辅助评估CC患者诊断和预后的潜在生物标志物。Objective To evaluate the association of tumor tissue long non-coding RNA Taurine Upregulated Gene 1(lncRNA TUG1)expression with diagnosis and prognostic in surgical patients with cervical cancer(CC).Methods 137 CC patients were enrolled in this prospective cohort study.The baseline demographic information,clinical and pathological features of all patients were recorded.The overall survival(OS)was calculated,and the lncRNA TUG1 expression in CC tumor tissues as well as adjacent tissues of patients were detected by Quantitative Polymerase Chain Reaction.Results lncRNA TUG1 relative expression was higher in CC tumor tissues sample than paired adjacent tissues sample(P<0.001).Moreover,CC tissue lncRNA TUG1 expression was positively correlated with International Federation of Gynecology and Obstetrics(FIGO)stage(P=0.006).Kaplan Meier curve and Log Rank test showed that tumor tissue lncRNA TUG1 higher expressions were correlated with worse OS(P<0.001)compared to lower expression.In addition,multivariate Cox's regression analysis revealed that tumor tissue lncRNA TUG1 high expression(P<0.001)was an independent factor for predicting worse OS.Tumor size≥4 cm(P<0.001),FIGOⅡstage(P<0.001),lymph node metastasis positive(P=0.001),and the cervical base infiltration depth≥2/3(P=0.002)were also independent factor for predicting shorter OS.Conclusion lncRNA TUG1 expression in cancer tissue can be used as a potential biomarker for diagnosis and prognosis of patients with CC.
