摘 要:目的:探讨小柴胡汤对完全弗氏佐剂(CFA)关节炎大鼠的作用机制。方法:将CFA关节炎大鼠随机分为正常组,模型组,小柴胡汤低、中、高剂量组,雷公藤多苷组,于造模后7d,正常组及模型组均予注射用水,小柴胡汤低、中、高剂量组分别予以小柴胡汤0.198、0.396、0.792g/mL,雷公藤多苷组予雷公藤多苷片混悬液0.21mg/mL,分别2mL/次,3次/d灌胃,连续给药28d后观察大鼠踝关节组织病理学及RT-PCR技术分析HIF-1α、VEGF mRNA的表达。结果:与模型组比较,各给药组病理评分显著减少(P<0.01);小柴胡汤中、高剂量组和雷公藤多苷组大鼠右后踝关节组织标本中HIF-1α、VEGF mRNA表达显著减少(P<0.05)。结论:小柴胡汤可以降低HIF-1α及下游靶基因VEGF的表达而改善滑膜炎症。Objective:To explore the mechanism of Xiaochaihu Decoction(XCHT)on CFA rats.Methods:CFA rats were randomly divided into the control group,the model group,the XCHT low,medium,high-dose group and the Tripterygium Glucosides group.On the 7th day after modeling,the drugs were given at 7d after the model was built.The rats were given to fill the stomach respectively three times a day,2mL/time.Both normal group and model group were given water for injection,XCHT low,medium,high-dose group were given XCHT 0.198,0.396,0.792g/mL,Tripterygium Glucosides group were given Tripterygium Glucosides tablet 0.21mg/mL.The expression of HIF-1α,VEGF mRNA was analyzed by RT-PCR and histopathology of ankle joint after continuous administration for 28 days.Results:Compared with the model group,the pathological scores of all administration each groups decreased significantly(P<0.01).The expression of HIF-1αand VEGF mRNA in the right posterior malleolus of rats in the XCHT medium,high-dose group,and the Tripterygium Glucosides group decreased(P<0.05).Conclusion:Xiaochaihu Decoction can reduce the expression of HIF-1αand downstream target gene VEGF and improve synovitis.
