摘  要:目的检测肝细胞肝癌(hepatocellular carcinoma,HCC)术后组织中微小RNA1288(microRNA-1288,miR-1228)的表达,探讨其与预后的意义,分析miR-1228与星形细胞上调基因1(astrocyte elevated gene-1,AEG-1)的关联性。方法选择57例HCC为研究对象,分别留取肿瘤组织和周边非肿瘤性肝组织作为观察组和对照组。应用实时荧光定量聚合酶链反应(quantitative real-time polymerase chain reaction,qPCR)法检测两组中miR-1228的表达。应用Western blot法检测观察组中AEG-1的表达,应用免疫组织化学(immunohistochemistry,IHC)方法检测观察组中增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)和凋亡因子Bcl-2相关X蛋白(Bcl-2 associated X protein,Bax)的表达,应用Spearman相关分析miR-1228与AEG-1、PCNA、BAX的相关性。结果观察组中miR-1228的表达明显高于对照组(P<0.05),观察组中miR-1228的表达在不同肿瘤直径、有无脉管累犯、有无转移和不同肿瘤分期(topography lymphnode metastasis,TNM)分期中差异有统计学意义(P<0.05),而在不同性别、年龄、分化程度、肿瘤部位中的差异无统计学意义(P>0.05)。miR-1228的表达与生存时间有关(P<0.05)。miR-1228与AEG-1(r=0.56,P<0.05)、miR-1228与PCNA(r=0.59,P<0.05)增殖指数均呈正相关性,miR-1228与BAX呈负相关性(r=-0.52,P<0.05)。结论HCC中miR-1228高表达是肿瘤形成的重要分子事件,参与病变的进展,AEG-1可能作为靶基因参与到miR-1228对肿瘤的调控作用中。miR-1228可能对细胞增殖和细胞凋亡有一定的调控作用。检测miR-1228的表达可能与HCC的预后有关。Objective To detect the expression of microRNA-1228(miR-1228) in hepatocellular carcinoma(HCC) tissues after operation, explore its prognostic significance, analyze the association with astrocyte elevated gene-1(AEG-1).Methods A total of 57 cases of HCC were selected as study subjects. Tumor tissues and peripheral normal liver tissues were taken as observation group and control group,respectively.The expression of miR-1228 in two groups were detected by quantitative real-time polymerase chain reaction(qPCR). The expression of AEG-1 in observation group was detected by Western blot method.The expression of proliferating cell nuclear antigen(PCNA) and Bcl-2 associated X protein(BAX) were detected by immunohistochemical(IHC) method in observation group. The correlation between miR-1228 and AEG-1, miR-1228 and PCNA, miR-1228 and BAX were analyzed by Spearman analysis.Results The expression of miR-1228 in observation group was significantly higher than that in control group(P<0.05).There were significantly differences of miR-1228 expression in different tumor diameters,vascular recidivism, metastasis and topography lymphnode metastasis(TNM) stages in observation group(P<0.05),no significantly differences were found in different gender, age, differentiation degree and tumor location(P<0.05). The expression of miR-1228 was related to survival time(P<0.05). Positive correlation was found between miR-1228 and AEG-1(r=0.56, P<0.05), miR-1228 and PCNA(r=0.59, P<0.05). Negative correlation was found between miR-1228 and BAX(r=-0.52, P<0.05).Conclusion The high expression of miR-1228 is an important molecular event in tumorigenesis of HCC, and it is involved in the progress of tumor. AEG-1 may be involved as a target gene in the tumor regulation of miR-1228. MiR-1228 may play a regulatory role in cell proliferation and apoptosis. Expression of miR-1228 may be related to the prognosis of HCC.
