摘 要:目的测定食物中唾液酸含量,探讨Neu5Gc摄入量与anti-Neu5Gc抗体浓度关系。方法采用液相色谱-质谱联用法测定常见食物中N-乙酰神经氨酸(N-acetylneuraminic acid,Neu5Ac)和N-羟乙酰神经氨酸(N-glycolylneuraminic acid,Neu5Gc)两种唾液酸(sialic acid,SA)的含量;随机抽取厦门市500名健康在校大学生并通过24h膳食回顾法及膳食史法调查膳食摄入情况,根据所测食物唾液酸含量计算Neu5Gc摄入量;用酶联免疫吸附试验(ELISA)检测被调查者anti-Neu5Gc抗体浓度,并用Spearman相关分析食物摄入量、Neu5Gc摄入量与anti-Neu5Gc抗体浓度相关关系。结果在检测食物中,奶类、蛋类、禽肉类、红肉类及水产品类等16种食物检测出了唾液酸;调查对象的膳食Neu5Gc摄入量呈正偏态分布,中位数水平为4.619 mg/d,四分位间距为6.408 mg/d;anti-Neu5Gc抗体水平为正偏态分布,中位数为3.072μg/ml,四分位间距为1.980μg/ml;Neu5Gc摄入量与anti-Neu5Gc抗体浓度之间存在相关性(rs=0.222,P<0.001)。结论红肉及奶制品的长期过多摄入可以引起Neu5Gc抗体的升高,可能增加患癌风险。Objective To determine the content of sialic acid in foods and explore the relationships between Neu5Gc intake and anti-Neu5Gc antibody concentrations.Methods The contents of N-acetylneuraminic acid(Neu5Ac)and N-glycolylneur-aminic acid(Neu5Gc)in foods were determined by liquid chromatography-mass spectrometry.Five hundred healthy college students in Xiamen were randomly selected,whose dietary intake was investigated by 24 h dietary recall method and food frequency method.Neu5Gc intake was calculated according to the measured sialic acid content.The concentration of anti-Neu5Gc antibody was detected by enzyme linked immunosorbent assay(ELISA),and the correlations between Neu5Gc intake/food intake and Neu5Gc antibody concentrations were analyzed by Spearman correlation analysis.Results Sialic acid was found in 16 kinds of foods,such as milk,eggs,poultry meat,red meat and aquatic products.The dietary Neu5Gc intake of the subjects showed a positive skewed distribution.The median value was 4.619 mg/d,and the Neu5Gc antibody level showed a positive skewed distribution.The median value was 3.072μg/ml.There was a significant correlation between Neu5Gc intake and the concentration of anti-Neu5Gc antibody(rs=0.222,P<0.001).Conclusion Long-term excessive intake of red meat and dairy products can lead to an increase in Neu5Gc antibodies,which may increase the risk of cancer.
