摘　　要：目的通过检测IL-9^+CD4^+T细胞及其相关因子在结直肠癌患者外周血中的表达,探讨其在结直肠癌发病机制中的作用。方法收集初次行手术治疗的60例结直肠癌患者的外周血,采用流式细胞技术检测IL-9^+CD4^+T细胞在结直肠癌患者及健康对照者外周血中表达差异,同时采用qRT-PCR方法检测PU.1、IL-9、CCL-2、CCL-5、CCL-20、CCL-27在结直肠癌患者的外周血中mRNA相对表达量。结果结直肠癌患者外周血中的IL-9^+CD4^+T细胞比例明显高于健康对照者(P<0.05),DukesA+B期患者的IL-9+CD4^+T细胞比例显著低于DukesC+D期(P<0.05);qRT-PCR结果显示IL-9、PU.1、CCL-2、CCL-5在结直肠癌患者外周血中的mRNA表达水平明显高于健康对照者(P<0.05)。结论IL-9^+CD4^+T细胞在结直肠癌患者外周血中比例增加,并与肿瘤的分期进展密切相关,可能与肿瘤微环境分泌的趋化因子作用密不可分。Objective To understand the role of IL-9^+CD4^+T cells in molecular pathogenesis of colorectal cancer.Methods Flow cytometry was employed to investigate the proportion of IL-9^+CD4^+T cells in in the peripheral blood monocyte from 60 colorectal cancer patients and healthy volunteers;while qRT-PCR was used to measure the relative expression level of PU.1,IL-9,CCL-2,CCL-5,CCL-20 and CCL-27.Results The ratio of IL-9^+CD4^+T cells in colorectal cancer patients was significantly higher than that in healthy controls(P<0.05);furthermore,IL-9+CD4^+T cells expression in Dukes Stage C or D patients was significantly higher than that in Dukes Stage A or B stage patients(P<0.05).According to qRT-PCR,the mRNA expression levels of IL-9,PU.1,CCL-2 and CCL-5 in colorectal cancer patients were significantly higher than those in healthy controls(P<0.05).Conclusion The increase in the relative prevalence of IL-9,CD4^+T cells may relate to the functions of chemotactic factors secreted by tumor microenvironment.
