摘  要:【目的】研究葡萄籽中多酚的最优提取工艺条件并测定其对脂肪酶的抑制率.【方法】通过响应面法优化葡萄籽多酚的提取工艺,然后分析不同体积分数乙醇对AB-8大孔吸附树脂纯化的葡萄籽多酚的脂肪酶抑制率.【结果】葡萄籽多酚最佳提取条件为70%乙醇、纤维素酶添加量为0.5 mg/mL、料液比1∶10、提取温度54℃、提取时间39 min、提取次数4次;得到的葡萄籽提取物对胰脂肪酶活性抑制活力IC50值为2.46 mg/mL.【结论】经过条件优化,多酚的提取率提高,所提取的多酚具有胰脂肪酶活性抑制活力.AB-8大孔吸附树脂法中不同乙醇洗脱组分对脂肪酶活力的抑制作用有一定差异,其中40%乙醇洗脱的葡萄籽多酚溶液对脂肪酶抑制效果最好.【Objective】 To optimize the conditions for extracting the polyphenols in grape seed and determine the lipase inhibition rate of extracted polyphenols.【Method】 The optimal process was obtained with response surface methodology.And the lipase inhibition rate of extracted polyphenols purified with AB-8 macroporous adsorption chromatography and eluted by ethanol solutions of different concentration was analyzed.【Result】 The results showed that the optimal extraction conditions were 70% ethanol,and the added amount of cellulase was 0.5 mg/mL,1∶10 solid-liquid ratio,extraction temperature 54 ℃,extraction time 39 min,extraction times 4 times.The IC50 value of lipase activity inhibition activity was 2.46 mg/mL.【Conclusion】 After the optimization,the extraction rate of polyphenols is increased,and the extracted polyphenols show inhibition activity on pancreatic lipase activity.In treatments of the AB-8 macroporous adsorption resin method,the inhibitory effects of different ethanol-eluting components on lipase activity are different,and 40% ethanol-eluted grape seed polyphenol solution has the best inhibitory effect on lipase.
