摘 要:目的检测白内障晶状体上皮细胞(LECs)中miR-126、caspase-3的表达情况,探讨二者在白内障晶状体上皮细胞凋亡中作用及表达关系。方法选取2017年5月至2018年6月收治的78例白内障患者为观察组,同期选取52例正常人(角膜移植术供体)为对照组,采用实时荧光PCR(qRT-PCR)法检测晶状体上皮细胞中miR-126、Caspase-3 mRNA表达水平;Western blot检测晶状体上皮细胞中Caspase-3蛋白表达水平;流式细胞仪测定细胞凋亡率;分析miR-126、caspase-3与晶状体上皮细胞凋亡率的相关性;分析miR-126与Caspase-3在白内障晶状体上皮细胞中表达的相关性。结果观察组白内障患者晶状体上皮细胞中miR-126、Caspase-3 mRNA及蛋白水平、晶状体上皮细胞凋亡率均高于对照组(P<0.05);白内障患者晶状体上皮细胞中miR-126、caspase-3表达与晶状体上皮细胞凋亡率呈显著正相关(r=0.669,P<0.05;r=0.633,P<0.05);白内障患者晶状体上皮细胞中miR-126水平与caspase-3表达水平呈显著正相关(r=0.599,P<0.05)。结论miR-126、caspase-3在白内障晶状体上皮细胞中高表达,二者与晶状体上皮细胞凋亡有关,提示二者可能通过影响晶状体上皮细胞凋亡参与白内障疾病发生及发展。Objective To investigate the expressions of miR-126 and Caspase-3 in lens epithelial cells(LECs)of patients with cataract,and to explore their effects on apoptosis of LECs.Methods A total of 78 patients with cataract who were admitted and treated from May 2017 to June 2018 were enrolled as observation group,at the same time,52 healthy subjects were enrolled as control group.The real-time fluorescent polymerase chain reaction(qRT-PCR)was used to detect the expression levels of miR-126,Caspase-3 mRNAs in lens epithelial cells,and Western Blot was used to detect the expression of Caspase-3 protein in lens epithelial cells,and the apoptosis rate was measured by flow cytometry.Moreover the correlation between miR-126,Caspase-3 and the apoptosis rate of lens epithelial cells was analyzed,and the correlation between the expression of miR-126 and Caspase-3 in cataract lens epithelial cells was analyzed.Results The expression levels of miR-126,Caspase-3 mRNAs and proteins and the apoptosis rate of lens epithelial cells in observation group were significantly higher than those in control group(P<0.05),and the expression levels of miR-126 and Caspase-3 in lens epithelial cells of patients with cataract were positively correlated with the apoptosis rate of lens epithelial cells(r=0.669,P<0.05;r=0.633,P<0.05),moreover,and the expression levels of miR-126 were positively correlated with those of Caspase-3 in lens epithelial cells of patients with cataract(r=0.599,P<0.05).Conclusion MiR-126 and Caspase-3 are highly expressed in cataract lens epithelial cells of patients with cataract,which are related to apoptosis of lens epithelial cells,it is suggested that the two factors may participate in the pathogenesis and development of cataract by regulating the apoptosis of affecting lens epithelial cell.
