摘 要:目的分析miR-27a对不同病变程度宫颈组织Siha细胞增殖的影响及表达水平差异性分析。方法选取宫颈组织病变患者120例,其中HPV-16阳性感染的正常宫颈组织患者为40例对照组,HPV-16阳性HSIL组织患者40例和HPV-16阳性早期宫颈鳞癌病变组织患者40例分别为研究组1、研究组2。采用逆转录-聚合酶链反应法检测三组患者宫颈组织中miR-27a表达水平,采用逆转录-聚合酶链反应法检测miR-27a转然后过表达及低表达时miR-27a的表达量;蛋白质印迹法实验技术检测Siha转然后SIPAIL3的表达水平。结果研究组1、2中miR-27a表达显著高于对照组,差异均有统计学意义(P<0.05);miR-27amimic组中mir-27a表达量显著高于miR-27amimic NC组,miR-27ainhibitor组mir-27a表达量显著低于miR-27a inhibitor NC组,差异均有统计学意义(P<0.05);miR-27amimic组Shia细胞增殖能力显著高于miR-27amimic NC组,miR-27a inhibitor组则弱于miR-27a inhibitor NC组,差异均有统计学意义(P<0.05)。结论miR-27a随着宫颈组织的病变程度加重,表达水平显著提升,且能够通过调控其下游靶基因SIPA1L3的表达促进Siha细胞的增殖并抑制其凋亡,进而促进了肿瘤的发生发展。Objective To analyze the effect of miR-27a on the proliferation of cervical tissue Siha cells and the difference in expression levels.Methods 120 patients with cervical tissue lesions were selected.Among them,40 patients with normal cervical tissue infected with HPV-16 positive infection were the control group,40 patients with HPV-16 positive HSIL tissue and 40 patients with HPV-16 positive early cervical squamous cell carcinoma tissue were study group 1,study group 2.Reverse transcription-polymerase chain reaction was used to detect the expression level of miR-27a in cervical tissues of the 3 groups of patients,and reverse transcription-polymerase chain reaction was used to detect the expression of miR-27a when miR-27a was transformed and then overexpressed and underexpressed;Western blot experimental technique was used to detect the expression level of Siha transfected SIPAIL3.Results The expression of miR-27a in study groups 1 and 2 was significantly higher than that of the control group,and the differences were statistically significant(P<0.05);the expression of mir-27a in miR-27amimic group was significantly higher than that of miR-27amimic NC group,miR-27ainhibitor The expression level of mir-27a in the group was significantly lower than that of miR-27a inhibitor NC,and the differences were statistically significant(P<0.05).The proliferation of Shia cells in miR-27amimic group was significantly higher than that of miR-27amimic NC group.Weaker than miR-27a inhibitor NC group,the differences were statistically significant(P<0.05).Conclusion As the degree of cervical lesion aggravates,the expression level of miR-27a is significantly increased,and it can promote the proliferation and inhibit the apoptosis of Siha cells by regulating the expression of SIPA1L3,which is the downstream target gene,thus promoting the occurrence and development of tumors.
