血清长链非编码RNA DANCR在非小细胞肺癌诊断中的临床应用

摘　　要：目的探讨长链非编码RNA DANCR在非小细胞肺癌(NSCLC)患者血清中的表达水平及其在NSCLC诊断中应用价值。方法收集82例NSCLC患者、45例肺部良性疾病患者及50例健康体检者的外周血标本。提取血清中的总RNA,应用实时荧光定量PCR技术检测血清中DANCR的表达水平,并分析其与NSCLC患者临床病理学特征之间的关系。结果DANCR在NSCLC患者血清的相对表达量为3.05(1.94,4.29),显著高于肺部良性疾病患者[1.33(0.96,2.48)]及健康对照者[1.01(0.81,1.50)](P<0.01),而肺部良性疾病患者与健康对照者之间差异无统计学意义(P>0.05)。NSCLC患者血清DANCR表达水平与TNM分期(P<0.05)及肿瘤大小(P<0.05)相关。ROC曲线分析结果显示:DANCR、CEA和Cyfra21-1的曲线下面积(AUC)分别为0.830、0.763、0.738。当联合DANCR、CEA和Cyfra21-1进行检测时,AUC达0.923,诊断敏感度和特异度分别为89.02%和84.00%。结论DANCR在NSCLC患者血清中表达水平升高,可能作为NSCLC辅助诊断的生物标志物。Objective To explore the relative expression of serum long non-coding RNA(lncRNA)differentiation antagonizing non-coding RNA(DANCR)in non-small lung cancer(NSCLC)patients and its clinical value.Methods The relative expression of serum lncRNA DANCR from 82 NSCLC patients,45 benign lung disease patients and 50 healthy controls were determined by RT-qPCR,and the correlations of DANCR expression level with pathological parameters of NSCLC were evaluated.Results The serum level of DANCR in NSCLC patients was significantly higher[3.05(1.94,4.29)],when they were compared with those in benign lung disease patients[1.33(0.96,2.48)]and healthy controls[1.01(0.81,1.50)](P<0.01).High DANCR expression in NSCLC patients were associated with tumor sizes(P<0.05),TNM stage(P<0.05).Areas under the ROC curve(AUC)of DANCR,CEA and Cyfra21-1 were 0.830,0.763 and 0.738.The combination of DANCR,CEA and Cyfra21-1 obtained the highest AUC,which was 0.923,the sensitivity and specificity were 89.02%and 84.00%,respectively.Conclusion Serum DANCR was increased in NSCLC patients and may be used as a molecular marker for the diagnosis of NSCLC.
