摘  要:目的研究CPT联合5-氟尿嘧啶(5-Fu)、奥沙利铂(L-OHP)对胃癌细胞的体外抑制作用。方法选取MGC823、SGC7901胃癌细胞株培养传代。两细胞系细胞分为空白对照组、CPT组、5-Fu组、L-OHP组、5-Fu+CPT组、L-OHP+CPT组。空白对照组不给予任何药物,CPT组给予CPT 1000μg/ml 2倍稀释直至0.004μg/ml;5-Fu组给予5-Fu 2.5×10^6 ng/ml 10倍稀释直至2.5×10^-4 ng/ml;L-OHP组给予L-OHP 1000μg/ml 2倍稀释直至0.002μg/ml;5-Fu+CPT组给予5-Fu阶梯浓度2.5×10^6 ng/ml 10倍稀释直至2.5×10^-8 ng/ml,并联合CPT 300 ng/ml给药;L-OHP+CPT组给予L-OHP阶梯浓度1000μg/ml 2倍稀释直至0.24μg/ml,并联合CPT 300 ng/ml给药。检测细胞在药物作用后的抑制效果。分析药物的半抑制浓度(IC50)和联合用药的药物协同作用。计量资料以均数±标准差(Mean±SD)表示,组间比较采用t检验。结果5-Fu+CPT组较5-Fu组抑制率显著提高,在MGC823、SGC7901细胞系差异均具有统计学意义(P=0.002、0.009)。L-OHP+CPT组较L-OHP组的抑制率也显著提高,在MGC823、SGC7901细胞系差异均具有统计学意义(P=0.037、0.040)。5-Fu+CPT组、L-OHP+CPT组较5-Fu组、L-OHP单药组IC50值显著降低,在MGC823细胞系、SGC7901细胞系差异均具有统计学意义(均P<0.001)。5-Fu与L-OHP分别与CPT联合作用于MGC823细胞系、SGC7901细胞系的存活分数低于70%采用Weeb系数计算的预估效应。结论5-Fu、L-OHP与CPT联合应用可明显提高对胃癌MGC823、SGC7901细胞系的抑制效果。5-Fu、L-OHP与CPT可产生良好的协同作用。Objective To study the inhibitory effect of CPT combined with 5-Fluorouracil(5-Fu),Oxaliplatin(L-OHP)on gastric carcinoma cells in vitro.Methods MGC823 and SGC7901 gastric carcinoma cell lines were selected for culture and passage.The two cell lines were distributed into blank control group,CPT group,5-Fu group,L-OHP group,5-Fu+CPT group and L-OHP+CPT group.The blank control group was not given any drugs,CPT group was given CPT 1000μg/ml 2-fold dilution until 0.004μg/ml;5-Fu group was given 5-Fu 2.5×10^6 ng/ml 10-fold dilution until 2.5×10^-4 ng/ml;L-OHP group was given L-OHP 1000μg/ml 2-fold dilution up to 0.002μg/ml;5-Fu+CPT group was given 5-Fu step concentration 2.5×10^6 ng/ml 10-fold dilution up to 2.5×10^-8 ng/ml,and combined with CPT 300 ng/ml;L-OHP+CPT group was given a 2-fold dilution of L-OHP step concentration 1000μg/ml up to 0.24μg/ml and combined with CPT 300 ng/ml.The inhibition effect of cells after drug action was detected.The 50%inhibitive concentration(IC50)of medicines and the synergistic effect of the combined drugs were analyzed.The measurement data were expressed as Mean±standard deviation(Mean±SD),and the t-test was used for comparison between groups.Results The inhibition rate of 5-Fu+CPT group was significantly higher than that of 5-Fu group,the differences in MGC823 and SGC7901 cell lines were statistically significant(P=0.002,0.009).The inhibition rate of L-OHP+CPT group was also significantly higher than that of L-OHP group,the differences in MGC823 and SGC7901 cell lines were statistically significant(P=0.037,0.040).Compared with 5-Fu+L-OHP group,IC50 value of 5-Fu+CPT group and L-OHP+CPT group decreased significantly,the differences in MGC823 and SGC7901 cell lines were statistically significant(all P<0.001).The survival fraction of 5-Fu and L-OHP combined with CPT on the two cell lines was lower than 70%,which was estimated by Weeb coefficient.Conclusions The combination of 5-Fu,L-OHP and CPT can significantly improved the inhibitory effect on MGC823 and SGC7901 cell lin
