摘  要:目的利用液相色谱-串联质谱法(LC-MS/MS)研究细菌性脑炎大鼠脑脊液代谢谱的改变,筛选与细菌性脑炎有关的潜在生物标志物,探寻相关代谢通路。方法将24只SD大鼠随机分为正常组(12只,颅内注射生理盐水)和感染组(12只,颅内注射金黄色葡萄球菌混悬液)。采用LC-MS/MS法,运用主成分分析(PCA)和偏最小二乘法判别分析(PLS-DA)分析比较感染组与正常组大鼠脑脊液的代谢谱差异,通过MetaboAnalyst 3.0分析相关代谢途径。结果两组大鼠脑脊液代谢产物相比,得到差异性代谢产物16种,其参与了丙氨酸、天门冬氨酸、谷氨酸代谢,甘氨酸、丝氨酸、苏氨酸代谢,氨酰基tRNA生物合成等代谢通路。结论本研究筛选出细菌性脑炎模型大鼠脑脊液中的16种差异代谢物,有助于解释颅脑细菌感染引起的代谢改变,有望为细菌性脑炎的早期诊治和机制研究提供帮助。Objective To use liquid chromatography-tandem mass spectrometry(LC-MS/MS) method to study the changes of cerebrospinal fluid metabolism in rats with bacterial encephalitis, to screen potential biomarkers related to bacterial encephalitis, and to explore related metabolic pathways.Methods Twenty-four SD rats were randomly divided into normal group(12, intracranial injection of physiological saline) and infection group(12, intracranial injection of S. aureus suspension). LC-MS/MS, principal component analysis(PCA) and partial least squares discriminant analysis(PLS-DA) were used to analyze and compare the metabolic profiles of cerebrospinal fluid in the two groups. MetaboAnalyst 3.0 was used to analyze related metabolic pathways.Results A total of 16 metabolites were found, which were involved in the metabolism of alanine, aspartic acid, glutamic acid, and biosynthesis of glycine, serine, threonine, aminoacyl tRNA.Conclusion This study screened out 16 metabolites in the cerebrospinal fluid of the rat model of bacterial encephalitis, which may help to explain the metabolic changes caused by bacterial infection of brain, and is expected to provide help for the early diagnosis and treatment of bacterial encephalitis.
