摘　　要：目的:研究转化生长因子β1(TGF-β1)在射线引起的骨髓间充质干细胞(BMSCs)电离辐射损伤中的作用及其相关分子机制。方法:建立细胞辐照模型,分别施加放射线、TGF-β1中和抗体、外源性TGF-β1、siNOX4等干预。并通过CCK8、蛋白印迹以及免疫荧光技术等方法检测细胞增殖、TGF-β1、NOX4、α-SMA含量。结果:放射线和外源性TGF-β1可造成BMSCs增殖能力下降、TGF-β1、NOX4、α-SMA合成显著增多(P<0.05);TGF-β1中和抗体可以明显抑制上述效应(P<0.05);siNOX4可部分抑制α-SMA的合成(P<0.05)。结论:放射线可以通过诱导TGF-β1合成进而刺激NOX4表达升高,促进BMSCs向肌成纤维细胞发生分化,参与形成了放射性颌骨骨髓炎的病理表现。Objective:To study the role of transforming growth factor-β1(TGF-β1)in radiation-induced injury of bone marrow mesenchymal stem cells(BMSCs)and its molecular mechanism.Methods:After establishement of cell irradiation model of BMSCs,radiation,TGF-β1 neutralizing antibody,exogenous TGF-β1 and siNOX4 were respectively applied to BMSCs.Cell proliferation,TGF-β1,NOX4 andα-SMA were detected by CCK8,Western blotting and immunofluorescence respectively.Results:Radiation and exogenous TGF-β1 decreased the proliferation of BMSCs while upgraded the synthesis of TGF-β1,NOX4,andα-SMA(P<0.05).TGF-β1 neutralizing antibody inhibited the above effects(P<0.05).siNOX4 inhibited the synthesis ofα-SMA(P<0.05).Conclusion:Radiation can upgrade the expression of NOX4 by inducing the synthesis of TGF-β1,which leads to the differentiation of BMSCs into myofibroblasts,and then forms the pathological manifestation of osteoradionecrosis.
