摘 要:目的评估酚苄明对阿尔茨海默病(AD)模型小鼠认知功能障碍的影响。方法20只C57BL/6雄性小鼠和20只3xTg(APP/Tau/PS1三转基因)雄性小鼠各随机分为2组(n=10)。一组为生理盐水对照组,另一组为酚苄明(PBZ)治疗组,分别标记为WT+NS组和WT+PBZ组、3xTg+NS组和3xTg+PBZ组。通过Morris水迷宫检测空间学习能力;ELISA测定海马组织可溶性Aβ水平;长时程增强(LTP)评估海马可塑性。结果与3xTg+NS组小鼠相比,3xTg+PBZ组小鼠逃避潜伏期显著缩短,在平台所在象限停留时间显著增加;3xTg+PBZ组在高频刺激后30min内fEPSP的斜率显著高于3xTg+NS组。WT+NS组和WT+PBZ组在高频刺激后30min内fEPSP的斜率差异无统计学意义。与3xTg+NS组小鼠相比,3xTg+PBZ组小鼠海马内可溶性Aβ显著减少(P<0.05)。结论酚苄明改善阿尔茨海默病模型小鼠的空间学习记忆功能,与减少海马组织内可溶性Aβ相关。Objective To evaluate the effect of phenoxybenzamine on learning and memory in Alzheimer’s disease model mice.Methods Twenty 4~6 months 3 xTg AD mice were randomly divided into phenoxybezamine injection(3 xTg+PBZ)group and saline injection(3 xTg+NS)group.Twenty wild type mice were divided into WT+PBZ and WT+NS groups.Morris water maze was used to evaluate the spatial learning ability,ELISA was used to measure the soluble Aβlevel in hippocampus,and hippocampal plasticity was assessed by long-term potentiation(LTP).Results Compared with 3 xTg+NS mice,the escape latency of 3 xTg+PBZ mice was significantly shortened,and the residence time at the platform quadrant was significantly increased.The slope of fEPSP in 3 xTg+PBZ group was significantly higher than that in 3 xTg+NS group within 30 minutes after high frequency stimulation,but with no significant difference for the slope of fEPSP between WT+NS group and WT+PBZ group within 30 minutes after high frequency stimulation.Compared with 3 xTg+NS group,soluble Aβin hippocampus of 3 xTg+PBZ group significantly decreased(P<0.05).Conclusion Phenoxybenzamine improved the learning and memory ability of Alzheimer’s disease model mice,which is related to reducing soluble Aβin hippocampus.
