摘 要:目的探讨富氢水减轻感染性休克大鼠小肠损伤的作用机制。方法SPF级SD大鼠40只,随机分为假手术组、感染性休克组、富氢水组及地塞米松组。于感染性休克发生后6 h处死大鼠,记录各组大鼠心率、平均动脉压;观察小肠病理损伤结果;酶联免疫吸附剂测定法(ELISA)检测肠组织中丙二醛(MDA)、超氧化物歧化酶(SOD)和二胺氧化酶(DAO)水平变化;检测大鼠肠组织中白介素-1β(IL-1β)、白介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)水平变化;应用Western blot检测大鼠肠组织中NF-κB和Nrf-2蛋白表达变化。结果富氢水及地塞米松能够平稳心率及升高血压,减轻感染性休克大鼠肠黏膜损伤,降低MDA水平并增加SOD及DAO活性,降低IL-1β、IL-6及TNF-α水平,抑制肠组织中NF-κB并促进Nrf-2蛋白表达。结论富氢水改善感染性休克大鼠肠损伤可能与调控NF-κB/Nrf-2表达相关。Objective To investigate the effect of hydrogen-rich water in reducing intestinal damage in septic shock rats.Methods Forty SPF SD rats were randomly divided into sham operation group,septic shock group,hydrogenrich water group(5 ml/kg)and dexamethasone group(10 mg/kg).The rats were sacrificed 6 h after septic shock.Heart rate and mean arterial pressure were recorded,pathological damage of small intestine was observed,malondialdehyde(MDA),superoxide dismutase(SOD)and diamine oxidase(DAO)levels in intestinal tissues were detected;levels of interleukin-1β(IL-1β),interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)in intestinal tissues,NF-κB and Nrf-2 protein expression were detected by Western blot.Results Hydrogen-rich water and dexamethasone stabilized heart rate and increased blood pressure,reduced intestinal mucosal injury in septic shock rats,decreased MDA levels and increased SOD and DAO activity,decreased IL-1β,IL-6 and TNF-αlevels,inhibited NF-κB in intestinal tissues and promoted Nrf-2 protein expression.Conclusion Hydrogen-rich water improved intestinal damage,which is related to the regulation of NF-κB/Nrf-2 expression in septic shock rats.
